Mycoplasma Pneumonia(MP) is one of major diseases which endanger sheep health.In recent years, China has become the hardest hit by MP which has caused enormous economic loss to the development of animal husbandry. Numerous studies have shown that miRNAs are related to the formation of tumor and the susceptibility and resistance to various diseases. Although the biological function of majority are not well understood, many studies have confirmed that the profiles of several miRNAs are heavily associated with several inflammatory lung diseases in human,mouse and rat. However, the related research has not been reported in sheep. On the basis of preliminary studies, this research project will screen some differential expressional miRNAs associated with MP from lung tissue of Chinese Merino Sheep. Then we will identify these miRNAs and confirm their function by using the methods of Northern blot,qRT-PCR,Western blot to clarify their mechanism on regulation and response to MP which will lay the foundation for further study on the correlation between the polymorphisms of miRNAs and target genes and MP. The implementation of this project has the important theory value and clinical significance for the prevention and treatment of MP and antidisease molecular breeding in sheep.
绵羊支原体肺炎是危害绵羊健康的主要疾病之一。近几年,我国已经成为了该病的重灾区,给畜牧业发展造成了巨大经济损失。大量研究证明,miRNAs与肿瘤的形成、多种疾病的易感性及抗病性相关。尽管对多数miRNAs的功能还不甚了解,但通过对人、小鼠、大鼠等体内miRNAs的研究,已经证实一些miRNAs的表达谱与几种肺部疾病高度相关,但在绵羊上的相关研究尚未见报道。本研究项目将在前期研究的基础上,从中国美利奴羊肺组织中筛选出一些与支原体肺炎相关的差异表达miRNAs,并通过Northern blot、qRT-PCR、Wwestern blot等方法对其进行鉴定和功能验证,以揭示miRNAs对绵羊支原体肺炎的调控和响应机制,为进一步研究相关miRNAs及靶基因多态性与绵羊支原体肺炎的相关性奠定基础。本项目的实施,对于绵羊支原体肺炎的防治和抗病分子育种工作具有重要的理论价值和临床指导意义。
