Phage clones that specifically binding with CD20 were selected form random phage displayed library using eukaryotically expressed CD20 fusion protein as ligand. The DNA of inserted peptides was sequenced after function identification of the positive phages, bioactive peptide binding with CD20 being gained. Further validation showed that these peptide can used to be coupled with detect reagents, toxins or drugs for detecting orientationally and targeting therapy, which may be of significant application perspective for basic research and drug development.
将CD20全长基因克隆到pIG1分泌型融合表达载体上,转染COS-7细胞,得到融合蛋白CD20-hIgG Fc;以些蛋白为配基,人噬菌体随机肽库中新和筛选到若干能与CD20特异性结合的阳性克隆;分析阳性噬菌体中的外源基因,将它们融合表达后再分别鉴定它们与CD20的结合活;进面合成活性多肽并鉴定其功能,为寻找用于诊断和导向治疗相关疾病的先导化合物奠定基础。
