Using clones from our laboratory and the consummate quality control system, We construct a high quality cDNA microarray platform, which has become an important tool for the research on medical genomics. 47 differentially expressed genes have been identified in the acute non-lymphocytic leukemia M2 subtype cell line HL-60 while comparing the gene expression profiles of HL-60 cells treated with all trans-retinoic acid at different timepoints using cDNA microarray, among which 19 genes are up-regulated while 28 genes are down-regulated. The differential expression of VCP and RBX1 genes involved in signal transduction indicates that NF-κB pathway plays an important role in the ATRA-induced differentiation. The results provide some valuable clues for the further research on the ATRA-induced differentiation network and the mechanisms of acute myelocytic leukemia.
用免疫亲和层析法分离纯化肾细胞中ADA和ADAvbp,经SDS-PAGE分析,得ADAbp分子量分别为70KD和110KD两条区带。此ADAbp有较弱的粮苷酶活性SOD活性,激活白细胞介素-2活性等功能。并得到ADAbp部分肽段的氨基酸序列。制备了抗ADA和抗ADAbp抗体,观察二者在细胞内的分布和表达性况,发现ADA和ADAvbp主要位于细胞膜上,且在肾癌细胞中ADAbp的表达量显著低于正常细胞。当腺苷浓度为50-150umol/L时,ADAbp对ADA的活性有较明显的抑制作用。还发现蛋白酶抑制剂磷酸二异丙基氟对ADAbp的DPPIV外肽酶活性有很强的抑制作用。上述结果有助于进一步探讨ADA和ADAbp在细胞分化,癌变过程中的作用机理,也为深入研究ADAbp的基因结构与表达调控奠定基础。
