Traditional Chinese Medicine Xingbi gelation nasal drops are new type of gel drop nose, which are based on the experience of the old Chinese Medicine, early basic research and clinical research showed that the effect is equivalent to Nasal glucocorticoid hormone,it can improve allergic rhinitis symptoms and nasal symptoms significantly. The domestic and foreign studies show that there is also a high airway reactivity in allergic rhinitis patients without asthma, Periostin and endoplasmic reticulum stress are related to the airway responsiveness. This project is intended to be through in vivo and in vitro experiments, to study on effects of Traditional Chinese Medicine Xingbi gelation nasal drops on the expression of Periostin,and the expression among of XBP-1, IRE1α belong to endoplasmic reticulum stress, to study on effects of allergic inflammation and airway hyper reaction in the upper and lower respiratory tract. The technology of laser scanning confocal scanning microscopy, Real-time, Western Blot, ELISA and other experimental techniques were used,the mechanism of Traditional Chinese Medicine Xingbi gelation nasal drops in the regulation of allergic rhinitis was discussed from different levels of RNA, protein level, cell and tissue morphology, and the same time, take Traditional Chinese Medicine Xingbi gelation nasal drops as the carrier to try to explore the local TCM Prevention and cure mechanism from the mechanism that early intervention of allergic rhinitis can delay or reverse the development of airway damage as a typical asthma.
醒鼻凝胶滴鼻剂是以名老中医经验方为基础研制的新型凝胶滴鼻剂,前期基础研究及临床研究均表明其与鼻用糖皮质激素效果相当,对过敏性鼻炎症状、鼻部体征均有明显改善作用。国内外研究表明,无哮喘的过敏性鼻炎患者亦具有气道高反应性,Periostin及内质网应激均与气道的高反应性有关。本项目拟通过体内和体外实验,研究醒鼻凝胶滴鼻剂对变应性鼻炎豚鼠鼻黏膜、肺组织Periostin,内质网应激XBP-1及IRE1α的表达,上下呼吸道变应性炎症及气道高反应等的影响,采用激光共聚焦扫描显微技术、Real-time PCR、Western Blot、ELISA等实验技术,从RNA、蛋白水平及细胞、组织形态学等不同层次探讨醒鼻凝胶滴鼻剂调控变应性鼻炎的作用机理,同时以“醒鼻凝胶滴鼻剂”为载体,试图探讨早期干预变应性鼻炎从而延缓或扭转下气道损伤发展为典型哮喘的局部中医药防治机制。
