Gui Yuan Jian effectively alleviated symptoms associated to decline in ovarian reserve (DOR) and up-regulated serum anti-Mullerian hormone (AMH) which directly refers to ovarian reserve function. Employing Immunohistochemical method, Western blot and RT-PCR, the project will firstly observe Gui Yuan Jian ameliorating AMH and AMHR expression in Granulosa Cells of all of follicals in ovary of cyclophosphamide model mice. Secondly, in vivo, the project will detect the expression and phosphorylation of AMH, AMHR, Smad2/3, Smad4, P21, P15, Rb protein, Cyclin B2, Cdc2 of AMH/Smads Signal Pathway in Ovarian Primary Granulosa Cells, to explain the mechanism of Gui Yuan Jian regulating proliferation of Granulosa Cells to inhibit follicular recruitment, and to explicate pharmacological mechanism of Gui Yuan Jian protecting ovarian reserve.
既往研究显示癸源煎能有效缓解卵巢储备功能下降(DOR)相关症状,并升调节反应卵巢储备功能的AMH水平。本项目运用免疫组化、Western blot、RT-PCR等检测方法进行研究,观察癸源煎对环磷酰胺模型小鼠卵巢各级卵泡颗粒细胞AMH、AMHR的改善作用;进一步通过体外实验研究“癸源煎”对初级颗粒细胞中AMH/Smads信号通路关键基因AMH、AMHR、Smad2/3、Smad4、P21、P15、Rb蛋白、Cyclin B2、Cdc2表达及其磷酸化,探索癸源煎降调节卵巢颗粒细胞增殖、抑制卵泡募集的作用机制,阐释癸源煎保护卵巢储备功能的机理,为中医药防治DOR的药理机制提供思路。
