QTL affecting callus regeneration ability in rice were analyzed using a F2.population from a cross combination between a low regeneration ability indica parent.Peiai64s and a high regeneration ability japonica cultuvar Joshu. Five QTLs.(QRg1-5、QCRg1-7a、QCRg1-7b、QCRg2-1、QCRg11-4)were detected and mapped.on chromosome 1,2 and 11. Variances explained by QTL were 1.11%、73.03%、.33.47%、80.79%、34.29% respectively. It is also found that the positive allele of.QCRg1-7a,QCRg11-4 was from Joshu..Three indica lines with high regeneration ability were obtained through.backcross breeding using Peiai64s as the recurrent parent and Joshu as the.non-recurrent parent. The regeneration rate of the lines was 71.9%,70.5%, 73.2% and.increased 66.5%、65.1% and 67.8% than Peiai64s respectively. This indicated that the.high regeneration genes detected is useful for improving the poor regeneration ability.in indica cultivars.
低再生力是生物技术在植物育种上应用的一大障碍,转移高再生力基因到农艺性状优良的遗传背景中是克服该障碍的唯一途径。本研究用RFLP、RAPD标记定位粳稻品种上州携带的二个控制高再生力的显性基因寻找与其紧密连锁的分子标记,分析其基因效应并培育籼型高再生力系统。为克隆该基因分析其结构功能以及育种上的应用奠定基础。
