Vaginal infectious disease is the most common gynecopathy and easy to repeated, since it is difficult to cure only by antibiotic therapy. And the function and mechanism of microRNA in mucosal infection and defense become hot topics. We have found and validated a microRNA named miR-1976, which was differentially expressed in vaginal mucosa between healthy women and vaginitis ones. It has been verified that miR-1976 also regulated the expression of integrin αvβ6 via CD105. In order to demonstrate that miR-1976 regulate the expression of integrin αvβ6 through CD105, leading to affecting the adhesion ability of vaginal epithelial cells and participating in the infection and defense mechanism of vaginal mucosa, the present research will study the expression characteristics and biological function of miR-1976 in vaginal epithelial cells, clarify the relationship among miR-1976, CD105 and integrin αvβ6, and evaluate the function and mechanism of miR-1976, CD105 and integrin αvβ6 in vaginal mucosa infection and defense. There were few reports about miR-1976 participating in vaginal mucosa infection and defense. Therefore, this research show certain creativity, and will provide new clues for controlling and preventing vagintis and looking for new targets for vagintis treatment. And this research will also help to provide ideas and clues for other mucosa research.
阴道感染性疾病为最常见妇科病,由于易反复发作而难以单纯靠抗菌药物治疗解决,因此探讨黏膜感染与防御的机制已成为阴道感染性疾病防治的重要方向。miR-1976是本课题组前期通过miRNA芯片筛选获得、差异低表达于阴道炎患者阴道黏膜组织的miRNA。前期研究发现miR-1976可能通过调节CD105的表达而调节整合素αvβ6的表达。本课题拟通过细胞生物学实验与动物实验,研究miR-1976在阴道上皮细胞中的表达特征与生物学功能,阐明miRNA-1976与CD105、整合素αvβ6间的关系,评估三者在阴道黏膜感染与防御中的功能与机制。miR-1976参与阴道黏膜感染与防御的研究未见报道,因此本研究具有源头创新性,将为阴道炎的防治提供新的靶标,也有助于为其他器官的黏膜感染与防御研究提供思路与线索。
阴道感染性疾病是最常见的妇科病。microRNA在黏膜感染与防御中的功能与机制是研究热点。miR-1976是本课题组发现并验证的一条差异表达于阴道炎与健康女性阴道黏膜组织的miRNA,并初步验证了其与可能作用靶标CD105的结合及对整合素αvβ6表达的调控作用。本课题通过老年健康与细菌性阴道炎女性阴道黏膜组织中miR-1976、CD105及整合素αvβ6的表达差异研究;阴道微环境相关因素对miR-1976在阴道上皮细胞中表达影响的研究;miR-1976在阴道上皮细胞中的生物学功能研究;miR-1976与CD105、整合素αvβ6在阴道黏膜感染与防御功能中的体内研究,系统探讨了miR-1976在阴道上皮细胞中的表达特征与生物学功能,阐明了miRNA-1976与CD105、整合素αvβ6间的关系,评估了三者在阴道黏膜感染与防御中的功能与机制,系统论证了“miRNA-1976以CD105为作用靶标,通过调节整合素αvβ6表达,影响阴道上皮细胞黏着能力,参与阴道黏膜的感染与防御机制”。miR-1976参与阴道黏膜感染与防御的研究未见报道,因此本研究具有源头创新性,可能为控制、预防阴道炎的发生以及寻找阴道炎治疗的新靶标提供新线索,也有助于为其他的黏膜感染与防御研究提供思路与线索。在本项目资助下,课题组已发表2篇中文文章、2篇SCI论文,另有2篇SCI论文正在撰写中。培养了3名硕士研究生、1名博士研究生;其中3名硕士研究生均已完成论文答辩、顺利毕业;1名博士研究生已完成毕业论文的大部分实验工作,计划于2019年完成毕业论文答辩工作。
