First, project had observed effects of extracts from Cichorii on hyperlipidemia and hyperuricemia and their companied disorders and studied on its mechanism. According to experiment results, the extracts lower TG, TC, UA of serum, reduce TC, NO and LPO of liver, and increase SOD activity, regulate blood viscosity, inhibit gene expression of xanthine oxidase in liver. It shows significant preventing and treating interacted disorder between lipid and purine metabolism. Mechanism is associated with extracts regulation enzyme activity and related gene expression. It has been found that the extracts be safety to function of liver and kidney. Second, project studied on pathology relationship of lipid and uric acid metabolism. With biochemistry and molecular biological technique the metabolic enzyme activity had been observed. It is found that gene expression and activity change of xanthine oxidase should be closely relation with lipid and purine interact disorder. Third, project studied on establishing hyperlipidemia complex hyperuricemia quail model which have similar way to human in lipid and uric acid metabolism and setting up hyperuricemia complex hyperlipidemia mice model. All pathological symptoms of models are similar with patient in clinic, so they are more suitable to pharmacology and pathology research.
探讨菊苣组分N1对血脂与尿酸代谢交互紊乱的影响及其机制。采用药理、生化、病理、分子生物学等多学科研究方法,选择不同的动物模型,从整体、细胞、受体与基因水平观察血脂与血尿酸代谢交互影响的因果关系及其分子生物学机理;阐明药物对高脂合并高尿酸血症的作用机理及对血管平滑肌细胞的保护作用。为临床寻找新的药物提供科学依据。
