The longicorn is one of the major forest boring-insect in China, and it is an elusive insect, which also has strong stress resistance and makes the common control measures relatively difficult and ineffective and caused devastating loss. This kind of insect has the habits of complementing nutrition and its adult living free and it has a long operating life. Through studying the adult host's selective mechanism, we can make use of the host's orient message to tentakill, or collect, the insect, and that is oneof the most important way to control the insect. The study will choose Batocera lineolata, which always caused serious harm in the southern part of China, as the experimental material. In this study we will detecte and analyze B. lineolata larval and adult's gene fragments to obtain the host selection relevanted gene, then we will compare the host's selectivity changes of adult through experience supplemental nutrition activities, afterwards we will analyze the host volatiles, finally we will measure the EAG and behavioral responses of B. lineolata to the host volatiles. We will analyse and compare B. lineolata's occurrence in the condition of the hosts, which have complemented nutrition, in the different distribution in the forest, and measure the enticing result, which make use of the host's semiochemical and factitious collocate the amount of tree bait, to clear and definite B. lineolata's heredity for the host's selectivity, obtain the chemical fingerprinting of the adult distinguish the host, expound the selective mechanism, obtain the relation between the host's, which have complemented nutrition, natural distribution and the population's change of B. lineolata, explore the the technology and method, which control B. lineolata's population by use the complemented nutrition host and it's volatiles.
天牛类蛀干害虫是我国森林的重大害虫,由于隐蔽生活,抗逆性强,因而防治十分困难,常造成毁灭性损失。这类害虫一般具有补充营养习性,成虫自由生活,历时较长,研究成虫寄主选择机制,利用寄主定向信息实施诱杀(集)是防治该类害虫的重要途径。本项目以我国南方发生普遍、危害严重的云斑天牛为研究对象,通过测定幼虫和成虫基因片段差异,检测寄主选择相关基因;测定经历补充营养活动后,成虫寄主选择性的变化;分析寄主挥发物成分,测定成虫对寄主挥发物成分的EAG和行为反应;研究比较林间补充营养寄主的不同分布状况下云斑天牛的发生情况;测定利用寄主信息化合物和林间人为配置诱饵树对成虫的引诱效果,从而明确云斑天牛寄主选择的遗传性和环间影响,破解成虫识别寄主的化学指纹图谱,揭示云斑天牛寄主选择机制;探讨林间补充营养寄主的自然分布状况与云斑天牛种群消长的关系;探索利用补充营养寄主及其信息化合物控制云斑天牛种群的方法和技术。
研究利用天牛对寄主植物的选择识别机制,开展种群持续控制越来越受到重视。项目针对云斑天牛幼虫为害寄主与成虫补充营养寄主常不同,且补充营养寄主种类多的特点,首先通过杨树云斑天牛不同虫态转录测序构建转录文库,分析不同虫态嗅觉基因差异;研究主要嗅觉基因在不同寄主来源云斑天牛及补充营养前后成虫间的差异,测定补充营养行为对识别寄主能力的影响,明确了云斑天牛寄主选择在嗅觉方面受遗传和化学环境影响。广泛测定分析补充营养寄主挥发物成分和含量,研究成虫行为与寄主挥发物日节律相关性,测定云斑天牛对主要挥发性成分单质和组合的电生理和行为反应,获得成虫识别寄主的化学信息物成分和比例。开展光信息在寄主选择中的作用研究,提出云斑天牛寄主定向是基于学化和光信息的复合信息系统。研究明确了云斑天牛在杨树和核桃林发生危害与补充营养寄主种类和分布的关系,提出并验证了定点保留或配置嗜食补充营养寄主,清除其余补充营养寄主防控云斑天牛的可行性。
