Jinchang in Gansu Province is known as China’s Capital of Nickel. Its nickel reserve ranks the first in China and the second in the world. Excessive residual of heavy metal nickel in the environment would affect oral microbial flora structure of the residents who live in the area. It is believed that Nickel pollution may be associated with periodontopathy. This project took Jinchang’s Nickel smelting zone as Nickel contaminated area in comparison with suburban agricultural zone as non-contaminated areas. Saliva samples were taken from residents in these two areas. The differences of the oral microbial flora structure were checked and compared between Nickel contaminated and non-contaminated areas. The project aims to reveal the effects of Nickel pollution on oral microbial flora structure. Periodontal disease epidemiology method was used to determine oral health condition and the incidence of periodontal disease. and analysis the difference. Spectrophotometric method was adopted to check the growth activity of Pg、Aa、Tf under gradient concentration of Nickel. Explore the relationship and mechanism between Nickel and oral microbial community and periodontopathy to provide the scientific basis for oral health and disease prevention in Nickel contaminated area.
甘肃省金昌市是我国著名的‘镍都’,镍储量居全国第一,世界第二。重金属镍在生活环境中的过量储集会影响居民口腔微生物的菌群结构,并与口腔牙周病的发生存在联系。本项目以金昌市镍冶炼区作为镍污染实验区,以金昌和张掖近郊农业区为第一、第二对照区,采集三个区域居民唾液样本,通过16s rRNA高通量测序,对比镍污染区与非污染区居民口腔微生物环境中菌群结构,研究镍污染对口腔微生态环境的影响;通过流行病学方法,检测比较两区域居民口腔卫生及牙周病发病情况,研究不同口腔菌群结构对口腔常见疾病发病率的影响;采用分光光度法测定在不同浓度镍离子的干预下,牙周病主要致病菌牙龈叶琳单胞菌(Pg)、伴放线放线杆菌(Aa)和福赛坦氏菌(Tf)的生长活性,探寻镍污染对口腔疾病发生的作用机制,为镍污染区口腔保健及疾病防治提供研究基础和科学依据。
