Hepatocellular carcinoma (HCC) is one of the common malignant tumor in China. Recurrence and metastasis is one of an important reason to poor prognosis. MMPs secreted by fibroblast, which induced by CD147, mediated tumor cell invasion, metastatic, proliferation and anti-apoptotic. However, the mechanism remains unclear. Our preliminary study found exosome produced by HCC cell can promote MMPs expression in fibroblast. But the signal pathway is unknown. We plan to adopt high-throughput technologies, such as RNA-seq, protein map to determine the role and mechanism of CD147-exsome-MMP in cross-talk of tumor cells and microenvironment. The results of the study would have a great significance to clarify the recurrence of liver cancer, and could provide some new theories and new targets to improve the curative effects of HCC.
肝细胞肝癌是我国常见的恶性肿瘤之一,复发转移是影响肝癌疗效的关键科学问题。成纤维细胞分泌的MMP是促进肝癌复发转移、增殖和抗凋亡的主要因素,其表达受肝癌细胞CD147的调控,但其机制不清。本课题组证实外泌体介导了肝癌细胞CD147诱导成纤维细胞MMP表达的过程,但具体机制尚不清楚。本课题拟通过RNA测序,蛋白质谱等技术,深入探索CD147-外泌体-MMP在改造肝癌微环境中的作用和机制。研究结果有助于深化肝癌复发转移的认识,也为提高肝癌治疗效果提供新理论、新方法、新靶点。
