质粒Pspt-hGH经酶切取得长约0.8Kb的hGHcDNA,逆转录病毒载体PDOR-Neo经酶解后与hGHcDNA连接形成PDOR-hGH重组逆转录病毒克隆。以Lipofectin介导将PDOR-hGH引入PA317细胞包装,形成感染性病毒颗粒PAC-hGH。小鼠成纤维细胞经传代,加入病毒液(PAC-hGH)孵育,并以G418筛选,直到出现抗性克隆(3T3-hGH)。扩增3T3-hGH提取其总DNA和RNA,分别以Southern印迹杂交和斑点杂交分析、签定、证实hGH基因已导入3T3细胞;免疫荧光法测定证实3T3-hGH培养液中hGH水平明显增高。经去垂体大鼠尾静脉注射已包装的感染性病毒颗粒PDOR-hGH,并以放射免疫法测定手术前后血清hGH水平,证实PDOR-hGH注射后血清hGH水平明显增高(P<0.01).
