Micro RNA participates in follicle development, ovulation and atresia by regulating the target gene and signal transduction pathway. Estrogen is an important pathophysiological factor of female ovarian dysfunction. Previous research findings show that moxibustion can effectively improve ovarian reserve function, and acupuncture intervention can up-regulate endometrial PI3K-Akt signaling pathway. Combined with domestic and foreign research progress, we suggest that the protective effect of moxibustion on ovarian function may be to improve the level of estrogen by regulating mi RNAs of the ovarian granulosa cells, and up-regulating the PI3K/Akt signaling pathway. In this study, the rat model of hypofunctional ovarian reserve will be established by oral administration of tripterygium wilfordii suspension. The protein and mRNA expression of PI3KP, p-Akt and Akt will be detected by Weston Blot and RT-PCR, to explore the protective mechanism of moxibustion on ovarian function; explore the mechanism of regulating mi RNAs in granulosa cells by second generation transcriptome sequencing. Based on PI3K/Akt signaling pathway, we studied the mechanism of regulation of moxibustion on ovarian function at transcriptional level, that in order to provide new ideas for the clinical application and mechanism study of moxibustion intervention in DOR.
miRNA通过对靶基因和信号转导通路的调节,参与卵泡发育、排卵和闭锁,影响激素分泌。雌激素是女性卵巢功能下降的重要病理生理因素,前期研究发现,艾灸可有效提高机体卵巢储备功能,且针灸干预可上调子宫内膜PI3K-Akt信号通路。结合国内外进展,我们提出“艾灸对卵巢功能的保护作用可能是通过调节卵巢颗粒细胞相关miRNAs,上调PI3K/Akt信号通路,从而改善机体雌激素水平,提高卵巢储备功能”的假说。本研究拟采用雷公藤悬浊液灌胃制作卵巢储备功能减退(DOR)大鼠模型,利用WB、RT-PCR技术检测卵巢PI3K、p-Akt、Akt蛋白和mRNA表达水平,探讨艾灸对卵巢功能保护的作用机制;利用二代转录组测序技术探索颗粒细胞miRNAs的调控机制。本研究基于PI3K/Akt信号通路,研究艾灸在转录水平对卵巢功能的调节机制,以期为艾灸干预DOR的临床应用及其机制研究提供新的思路。
课题组前期临床及动物实验研究发现,艾灸在提高机体E2、AMH等水平方面有肯定疗效,且针灸干预可上调子宫内膜PI3K-Akt信号通路。结合国内外进展,本研究采用雷公藤悬浊液灌胃制作卵巢储备功能减退(DOR)大鼠模型,并进行艾灸干预,观察其动情周期、血清性激素变化,利用HE染色观察卵巢组织形态学变化, qRT-PCR及WB技术检测卵巢PI3K、p-Akt、Akt蛋白、mRNA表达水平,评价艾灸对卵巢功能保护的作用机制。研究发现,艾灸能改善紊乱的动情周期,促进卵泡生长,减少闭锁卵泡数量,提高血清E2、AMH水平,降低血清FSH、LH水平,艾灸后PI3K/AKT信号通路被激活,说明艾灸可减轻TGs诱导的卵巢功能下降,调节激素水平,其机制可能与PI3K/AKT信号通路有关。利用二代转录组测序技术筛选卵巢颗粒细胞中mi RNAs的差异表达,利用生物信息学方法,分析艾灸干预DOR差异表达基因的功能及网络;并应用RT-PCR技术,对筛选出的具有已知生物学意义的靶点基因mRNA及蛋白表达进行检测,发现艾灸可能是通过下调rno-miR-92b-3p、mdo-miR-26-5p_R+1_1ss10TC、rno-miR-206-3p、pal-miR-9993b-3p_1ss6GA、bta-miR-7857-3p_R-1、rno-miR-219a-2-3p_1ss10GC、mmu-mir-3968-p5_1ss10AT、PC-5p-6478_1795,上调rno-miR-664-2-5p_R+1保护DOR,以探讨艾灸在基因水平发挥整体调节作用的特点,揭示艾灸改善DOR可能的分子作用机制,以期为艾灸在DOR的临床应用提供可靠的科学依据,也为进一步研究艾灸对DOR的分子干预机制提供新的研究方向和研究思路。
