A high efficient transformate system of Populus detoides x P..simonii had been developed. Modified Bt CryIA gene and cowpea trypsin.inhibitor (CpTI) were co-ransforted into NL-10806. PCR and.Southernblot analysis showed that Bt and CpTI genes had been.intergrated into 9 clones. Insect bioassay indicated that transgenic.plants of BC55 and BC5 were significantly resistant to the larvae of.Lymantria dispar. Transgenic clones were planted in the way of random.area arry and the results of two times of insects resistant tests.showed that transgenic clones resistanted to insects.
利用DNA杂交技术、RNA杂交技术、mRNA差异显示技术及酶联免疫吸附测定等现代分子生物学检测手段,研究杨树转双价抗虫基因植株在生长发育过程中外源基因的表达;利用甲基化测定技术,研究甲基化对外源基因表达的影响。本项目首次对外源基因能否在林木较长的生长周期中稳定表达进行研究,为深入了解外源基因沉默机制,改进林木转化技术提供依据。.
