An insect neurotoxin gene from the scorpion Buthus martensi Karsch, cloned by our group, was constructed into a plant transformation vector together with an insect specific chitinase gene. Agrobacterium mediated transformation approach was employed to obtain highly and endurable insect resistant transgenic crop cultivars. These two insect specific genes were also introduced into the genome of a multiple nuclear polyhedrosis baculovirus, Aotographa californica (AcMNPV). Bioassay showed that the recombinant AcMNPV was superior to the wild virus in killing cotton bollworm (Helicoverpa armigera). During the period of conducting the project, 8 papers were published, 1 international and 3 national symposiums were participated, and 5 Msc students graduated. The project was also approved as an international collaboration project with Australia.
本研究利用从我国蝎种—东亚钳蝎中克隆到杀虫效果好的挛缩型蝎毒蛋白基因与昆虫特异性几丁质酶基因连接,构建双价抗虫基因,并采用不同的启动子调控该基因在转化植物中的表达,以创造转化频率高、转化后表达效果好的植物转基因载体系统,可作为进一步开展作物高级抗虫转基因育种研究奠定良好的基础。.
