Wampee fruits contain significantly high levels of flavonoids and thus exhibit high antioxidant activity. Therefore they are potential resources for developing antioxidant-rich food. However, little researches have been done regarding the differences in total flavonoids content, major flavonoid components and their relationship to total antioxidant activity of different wampee genetic materials, which has hampered the efficient utilization of wampee resources. In this study, fruits of the 150 accessions preserved in the National (Guangzhou) Wampee Germplasm Repository will be extracted and analyzed for their total flavonoids. HPLC-MS will be used to analyze wampee flavonoid profiles to identify their monomer components. Total flavonoids from each accession will be measured for their total antixidant activities by using the ORAC method and the cell antioxidant activity (CAA) assay. The correlations between total flavonoid content and total antioxidant activity will be analyzed, and similar analysis will also performed between each of the major flavonoid components and total antioxidant activity, for each wampee accession, to understand the relative contribution of the total flavonoids and the individual flavonoid components to the overall antioxidant activities. The major antioxidant contributor(s) will be used to identify potential wampee accessions that will be possibly used in direct production of antioxidant food and in breeding for high antioxidant wampee cultivars.
黄皮果实富含黄酮类化合物,具有较高的抗氧化活性和潜在的开发抗氧化功能食品的价值,但目前关于不同黄皮种质果实中黄酮类化合物种类、含量及抗氧化活性还缺乏系统的研究,致使相关开发利用进展缓慢。本研究拟以“农业部广州黄皮种质资源圃”保存的150份不同黄皮种质的果实为材料,提取总黄酮,测定总含量;用HPLC-MS法鉴定主要黄酮单体化合物;分别用ORAC法和细胞抗氧化活性评价方法(CAA法)分析总黄酮提取物的抗氧化活性;分析总抗氧化活性与总黄酮含量、主要黄酮单体含量之间的相关性,阐明总黄酮含量、主要黄酮单体含量对抗氧化活性的相对贡献率,据此鉴别有用资源,为直接开发抗氧化食品或用于高抗氧化黄皮育种奠定基础。
黄皮作为药食同源的水果,富含黄酮类化合物,具有较高的抗氧化活性及保健功效。本项目通过对“农业部广州黄皮种质资源圃”中的151份不同种质黄皮果实总黄酮含量、单体成分以及抗氧化活性的评价,筛选出具有高类黄酮含量以及高抗氧化活性的种质5份;结果显示,不同种质黄皮果实中总黄酮含量差异较大,最高值为909.9 mg CE/ 100g FW(鸡心黄皮),最低值为49.15 mg CE/ 100g FW(从城甜黄皮);不同种质黄皮果实的类黄酮单体成份主要为芸香叶苷(含量范围为10~30 mg/100g FW);黄皮果实的抗氧化活性与总黄酮含量呈正相关关系,总黄酮含量低的甜黄皮类抗氧化活性较低,而总黄酮含量高的酸黄皮类抗氧化活性较高;黄皮提取物对人肝癌细胞(HepG2)具有一定的细胞毒性,酸黄皮类的抗增殖活性较高,而甜黄皮较低;综上所述,本研究表明鸡心黄皮等酸黄皮类具有较好的开发应用前景。
