Correspondence between prescription and syndrome is core and basic principle in the TCM research。A shackles or bottleneck of basic research is the lack of an animal model to reflect the characteristics of clinical syndromes. Studies of specific model corresponding to the prescription is nearly blank. Based on our previous work and the principle of correspondence between prescription and syndrome, this study intends to characterize and simulate the biological effects of the serum from patients with Guizhi Tang syndrome in vivo and in vitro:Human lung macrophages(AM cell line) were be treat with both serum from patients with Guizhi Tang syndrome(or other exterior syndrome )/or different ratios of poly(I:C)-LPS and Guizhi Tang-containing serum(or other diaphoretic recipe),the simulation and the interference effects (TLR-3,4,6,9 and TNF, IL-6, IFN etc. ) were evaluated with principal component analysis and partial least squares method,the poly(I:C)-LPS ratios with best stimulation and simulation with the serum from patients with Guizhi Tang syndrome /best interference by Guizhi Tang-containing serum were used in rats,and more than 10 conscious physiological indicators were measured simultaneously to evaluate the simulation and the interference effects, as like above. All above will be done with the wish of providing further experimental support for revealing the pharmacological action of Guizhi Tang and the characteristics Guizhi Tang syndrome,and providing the new method and valuable ideas to studies alike.
方证相应是中医药临床遣药用方和基础研究的基本思维和基本原则,基础研究的一个桎梏或瓶颈是缺少反映证侯临床特质的证侯动物模型,而具体方剂相对应的汤证模型研究几乎处于空白状态。基于方证相应原则和课题组前期工作,本课题拟对桂枝汤证患者血清在细胞和动物进行表征和模拟:以不同比例poly(I:C)-LPS组合以及桂枝汤证患者血清刺激人肺巨噬细胞,桂枝汤含药血清干预,利用主成份分析和偏二小最乘法综合分析并比较(TLR-3、4、6、9以及TNF、IL-6、IFN等)的模拟和干预效果,选择模拟度最高和/获干预效果最好的poly(I:C)-LPS组合刺激大鼠并观测10多项清醒自由同步测定的生理指标,同上法分析刺激模拟度以及桂枝汤的干预效应,旨在尝试探索建立桂枝汤“汤证模型”,为进一步更好的揭示经方桂枝汤功效以及其对应证——太阳中风表虚证的现代科学内涵服务,同时也为其他汤证研究提供可资参考的方法和思路。
方证相应是中医药临床遣药用方和基础研究的基本思维和根本原则,基础研究的一个桎梏或瓶颈是缺少反映证侯临床特质的证候动物模型,而具体方剂相对应的汤证模型研究几乎处于空白状态。基于方证相应原则和课题组前期工作,本课题完成了如下工作:比较不同比例poly(I:C)-LPS组合刺激大鼠肺巨噬细胞和大鼠的综合效应(TLR-3、4、2、6以及TNF、IL-1等)以及桂枝汤(或其含药血清)的干预作用针对性,模拟桂枝汤证患者血清生物效应,尝试建立了第一个汤证模型:桂枝汤证体外细胞模型和大鼠桂枝汤证模型(后者有待进一步确证),该研究为进一步揭示经方桂枝汤功效以及其对应证——太阳中风表虚证的现代科学内涵提供了更好的针对性强的基础研究载体,同时也为其他汤证研究提供可资参考的方法和思路。
