Chicken infectious anemia (CIA) is caused by Chicken infectious anemia virus (CIAV), which causes destruction of erythroblastoid cells in the bone marrow and lymphoid depletion of the thymic cortex in susceptible chicks. CIAV can transmit vertically through the eggs of infected breeder flocks resulting in a severe anemia and trophy of the lymphoid organs. Also the virus can be transmitted horizontally when maternal antibody was disappeared, resulting in a sub-clinical infection that adversely affects growth and profitability in broiler flicks. CIAV has recently been noticed and considered to be one of the most important avian diseases. However, the methods used to control CIA now are not effective..In this study, we have amplified the genes of vp1 and vp2 of CIAV by PCR, and inserted them into genome DNA of HVT(Herpesvirus of Turkey) to construct recombinant HVT, which can express these two types of proteins. Also, we have made the research in the expression and immunogenicity of vp1 and vp2 of the recombinant virus. Our research contents completed include as follow:1.Sequencing and analyzing of encoding sequences of CIAV M9905 strain.2.Constructing 3 recombinant plasmid pCISIvp1-vp2,pCIvp1,pSIvp2 as DNA vaccine and comparing the protection of them in SPF chickens with inactive vaccine CUX-1.3 Expression of VP1 and VP2 genes of CIAV by baculorvirus expression systems4.Immunogenicity determination of VP1 and VP2 proteins of CIAV.5..Construction of a turkey herpesvirus recombinant containing VP1 and VP2 genes of CIAV.6.Immunogenicity determination of a turkey herpesvirus recombinant containing VP1 and VP2 genes of CIAV. Study on this project is laying a foundation of the research and development of effective CIA vaccine.
鸡传染性贫血病是鸡最重要的免疫抑制病之一,目前还没有安全有效的疫苗对该病进行预防。本研究拟采用PCR方法扩增和克隆鸡贫血病毒的VP1和VP2基因,并将其插入火鸡疱疹病毒℉VT)DNA中,构建表达这两种蛋白的重组HVT;同时对VP1和VP2在重组病毒中的表达及其免疫原性进行研究。为安全有效的鸡传染性贫血病疫苗的研制和开发奠定基础。
