辐射对肝P450酶的影响及人参干预作用研究

Cytochrome P450 oxidase plays an important for many endogenous and exogenous compounds biological conversion in vivo. Many diseases，such as cancer, cirrhosis，relate to function changes of hepatic P450 enzyme.According to literature reports,radiation can cause changes in hepatic P450 enzyme.but its law and correlation with irradiation dose are not clear. Be on the basis of ginseng against radiation damage and previous work of ginseng regulating P450 enzyme,in this project we will be ready to stuy the influence of different doses of radiation on hepatic P450 enzymes from the gene, protein and enzyme activity levels,and explore radiation-sensitive P450 enzyme(subtypes),and research ginseng regulating hepatic P450 enzymes under the radiation conditions,and analyze the chief effective components of ginseng. The completion of the project would contribute to clarify the correlation between different doses of radiation and the changes of P450 enzyme，provide new ideas for studying natural medicine resistance to radiation damage.It also could provide the experimental basis for interpreting the correlation between hepatic P450 enzymes and radiation induced Shang-Jin-Hao-Qi,and ginseng's effect on Bu-Qi-Sheng-Jin.

细胞色素P450氧化酶对许多内源、外源性化合物在体内的生物转化有重要作用，肿瘤、肝硬化等众多疾病与肝P450酶功能改变有关。文献报道辐射可引起肝P450酶的变化，但其变化规律及与照射剂量的相关性尚不明确。本研究在人参抗辐射损伤作用、以及前期人参调节P450酶的工作基础上,从基因、蛋白及酶活性水平探讨不同照射剂量对肝P450酶的影响，明确辐射敏感P450酶（亚酶），同时考察人参对辐射条件下肝P450酶的调控作用，并分析其有效物质基础。本课题的完成将有助于探明不同照射剂量辐射损伤与P450酶的相关性，为天然药物抗辐射损伤作用研究提供新的思路；也为阐释肝P450酶与辐射致伤津耗气的关系，人参补气生津的生物学基础提供实验依据。

本实验以大鼠肝P450酶辐射损伤为研究对象，探究不同照射剂量和时间点下辐射对大鼠肝P450酶的影响及人参的干预作用。研究结果显示：辐射能引起P450酶活性的改变，不同的亚酶对辐射的敏感程度不一样，受辐射影响后变化不一样,；大鼠CYP1A2、2B1较CYP2E1、3A4更容易发生辐射损伤；人参总皂苷对辐射大鼠P450同工酶（CYP1A2、CYP2B1、CYP2E1和CYP3A4）的损伤能起到较好的修复效果。本研究在一定程度上揭示辐射对P450酶的影响和相关亚酶的自身变化情况，以及人参对辐射损伤的干预效果，为辐射防护研究和抗辐射天然药物开发提供了实验数据支撑。
