Uveitis is a recurrent autoimmune eye disease harming vision health severely, and at present clinic treatments lack efficient and harmfulless medicine to avoid its recurrence. YanYanKang granule (patent number: 201210441467) developed by our project under the theoretical guidance of integration concept of traditional Chinese medicine has independent intellectual property rights and definite effects, but its mechanism is unclear. Gamma/delta T cells induce acquired immune response by regulating Th1 Th17 cells and plays an important role in the occurance of experimental autoimmune uveitis (EAU). Our project will establish EAU model and tEAU model, and use YanYanKang granule for intervention ,to detect the influence of proliferation and differentiation of Th1 and Th17 cells and regulatory mechanisms of turning pathogenic immune cells into immune cells by gamma/delta T cells,by means of pathology、immunohistochemical、western blot、flow cytometry、real-time quantitative PCR techniques on the organic、cellular and molecular levels, to discuss the key mechanism of the treatment of YanYanKang granule on uveitis ,laying theoretical foundation for clinical application of YanYanKang granule , pioneering a new way for traditional Chinese medicine treatment of uveitis.
葡萄膜炎是一种严重损害视觉健康的反复发作的自身免疫性眼病,目前临床缺乏疗效高毒副作用小的治疗药物。本课题组在中医整体观念理论指导下研制的眼炎康颗粒(专利:CN201210441467)具有独立知识产权,临床疗效确切,但其作用机制尚不清楚。gamma/deltaT细胞通过调节Th1、Th17细胞诱导获得性免疫反应,在实验性自身免疫性葡萄膜炎(EAU)发病中具有关键作用。本课题拟建立EAU模型及tEAU模型,采用组织病理、免疫组化、Western blot、流式细胞术、实时定量PCR等技术从器官、组织、细胞及分子等多层次水平检测眼炎康颗粒干预Th1和Th17细胞增殖、分化的影响及gamma/delta T等调节性免疫细胞(Treg)的调节机理,从免疫学角度探讨眼炎康颗粒治疗葡萄膜炎的关键机制,为眼炎康颗粒的推广应用奠定理论基础,为中医药整体治疗葡萄膜炎开创一条新途径。
