Essential hypertension is a kind of complicate multifactorial disease,and its development is determined by a combination of genetic susceptibility and environmental factors. Hypertension is the most common factor of vascular endothelial damage. Vascular endothelial dysfunction will increase the occurrence and development of hypertension. miRNA can be influenced the occurrence of hypertension through regulate the function of vascular endothelial cell. Prevention and treatment of hypertension has been hard work of cardiovascular researcher. Angelica is genuine medicinal herbs in Gansu which has the effect of promoting blood circulation and removing blood stasis. Our group of the previous findings that differences in miR122 gene expression after treatment with Angelica and has been verified.This study will select SHR and HUVEC induced by AngⅡas research materials. Analysis miR122 and its target genes and related signaling pathways in vascular endothelial system and explore the mechanism of Angelica treatment, the intervention mechanism of regulation of miR122 on endothelial cells and Angelica on vascular endothelial system, clarify the influence miR122 on the endothelial system, explore the effect of Angelica on miR122, on its target gene SLC7A1 and related signal pathway; through the above research to reveal the effect of miR122 on target gene and regulation mechanism against cardiovascular disease.Reveal the effect of protect mechanism of Angelica sinensis on the vascular endothelial system, which will not only provide theoretical basis of studying Gansu geoherbs, but also apply new drug targets for future RNA interference therapy of hypertension.
原发性高血压是是多因素所致的疾病,其中血管内皮功能障碍是高血压发病的重要机制之一,而miRNA可通过调控血管内皮细胞的功能影响高血压的发生、发展。当归是甘肃道地药材,有关当归对高血压miRNA表达谱的影响本科研小组已做了相关研究,并证实miR122为当归处理后血管组织的差异表达基因。本研究拟以HUVEC为实验材料,通过双荧光素酶报告基因检测系统明确miR122调控SLC7A1基因对血管内皮细胞表达的影响;同时以AngⅡ诱导损伤的HUVEC及SHR为研究材料,通过细胞与动物实验分析当归对高血压血管内皮系统miR122及其靶基因和相关信号通路及血管活性物质的影响,以此揭示miR122对血管内皮的调控机制及当归对高血压血管内皮系统的干预机制,阐明miR122对高血压血管内皮系统功能的影响以及当归对其的保护作用,为更深入研究甘肃道地药材提供理论和实验依据。
原发性高血压一种常见的多因素所致的疾病,作为心血管事件的重要危险因素,常引起严重的心、脑、肾并发症。而血管内皮功能紊乱是高血压发病的重要病理基础,内皮功能紊乱使内皮细胞分泌舒血管和缩血管活性因子失衡,最终导致一系列心血管事件。microRNA参与高血压的发生、发展和结局。miRNA 通过调控与内皮细胞功能密切相关的基因表达而参与内皮细胞和血管功能的调节,从而影响高血压的发生。当归是甘肃的道地药材,尤以甘肃岷县当归品质最佳。研究已证实当归对血压具有干预作用。本实验小组前期关于自发性高血压大鼠SHR的miRNA基因芯片的研究结果发现miR-122为当归处理后血管组织的差异表达基因,通过荧光定量PCR验证后也得以证实,这为本次研究的开展提供了科学依据。本次研究以自发性高血压大鼠及AngⅡ诱导损伤的HUVECs细胞为研究对象,通过在体与离体实验分析当归对高血压血管内皮活性物质及miR122及其miR122靶基因及相关信号通路表达的影响,研究结果证实miR122对SLC7A1具有调控作用,当归对血管内皮具有保护作用,可以提高AngⅡ诱导损伤的HUVECs细胞活力,并不同程度地降低血清中EMPs、vWF、ET-1、VEGF水平,提高NO的表达量。在细胞与动物实验中均证实当归干预后miR122的表达量降低,以当归高剂量组为著,SLC7A1的 mRNA及蛋白表达量均不同程度有所提高,同时当归挥发油能调节PI3K/Akt/eNOS信号通路蛋白表达,并呈现一定的量效相关性,在细胞与动物研究中所得结果基本一致。本项研究表明当归对高血压血管内皮具有一定的保护作用,可能通过下调miR-122负向调控SLC7A1的表达,激活PI3K-AKT-eNOS信号通路,对血管内皮发挥保护作用。本项研究从分子层面阐明了当归对血压的干预机制,为我省道地中药材的药理研究奠定实验基础,也为今后高血压的新药研发及mi RNA干扰治疗高血压提供新的靶点。
