Alzheimer's disease ( AD ) has become one of the main death diseases in the elderly, the clinical therapy of AD remains the world's problem. In recent years, the development of stem cell and its related technology make a spurt of progress, which provides an opportunity on study the therapy of AD. But stem cell function ( including migration, proliferation, differentiation and matrix secretion ) regulation depends on signal molecules to achieve, in previous work the applicant discussed the protective fuctions of two kinds of growth factor, glial cell line-derived neurotrophic factor ( GDNF ) and hepatocyte growth factor ( HGF ) in cerebral infarction model, this study is based on previous research, dicuss of GDNF, HGF and MicroRNA-124 in bone marrow stromal cell ( BMSCs ) proliferation in vitro and to the regulation and mechanism of neural cell differentiation. In mouse AD model, observe therapeutic effects of the modified BMSCs through the observation of nerve cell death and AD related protein changes. Further analysis nerve cell death mechanism of AD, provide new theoretical basis for the therapy of AD.
阿尔茨海默病(AD)已成为老年人的主要致死疾病之一,目前临床治疗AD仍是世界难题。近年来,随着干细胞及其相关技术的发展突飞猛进,这为AD的治疗研究提供了契机。但干细胞的功能(包括迁移、增殖、分化和基质分泌)都有赖于信号分子的调控来实现,在前期工作中申请人探讨了两种生长因子胶质细胞源性神经营养因子(GDNF)和肝细胞生长因子(HGF)在脑梗死模型中的神经保护作用,本研究是在前期研究基础上,在体外探讨GDNF、HGF以及MicroRNA-124对骨髓基质干细胞(BMSCs)增殖和向神经细胞分化的调控作用及机制,在AD小鼠模型中,通过观察海马及额叶神经细胞死亡及AD相关蛋白表达变化,探讨修饰后BMSCs的干预作用。进一步解析AD神经细胞死亡机理,为AD的治疗提供新的理论基础。
虽然在C9orf72肌萎缩性侧索硬化(C9-ALS)患者中证明了核质运输(neucleocytoplasmic transport, NCT)蛋白的异常分布,但是难以确定这种变化是否也与其他ALS类型疾病相关。在本研究中,我们调查人类Cu / Zn超氧化物歧化酶-1突变转基因(SOD1-Tg)小鼠和散发性ALS患者RanGAP1和4代表性核质运输蛋白(GP210,NUP205,NUP107和NUP50)的时空表达和共定位关系。与野生型(WT)相比,这些NCT蛋白在10至18周的SOD1-Tg小鼠中显示年龄依赖性和进行性核沉淀,以及腰脊髓运动神经元中的细胞质异常表达。双免疫荧光分析显示RanGAP1/NUP205和NUP205/NUP107的异常核共定位,同时,GP210/NUP205主要共定位在运动神经元的核包膜中。此外,RanGAP1,GP210和NUP50在SOD1-Tg小鼠和ALS患者中显示类似的异常核沉淀和细胞质的表达上调,此外,在本研究中也观察到RanGAP1/TDP-43和NUP205/TDP-43的共定位。本研究表明,RanGAP1和4种核质运输的功能障碍可能在SOD1-Tg小鼠和人散发性ALS患者的发病相关,这些功能障碍可能是ALS的一个基本途径,同时核质运输蛋白可以适于药物治疗干预的靶点。
