Wu-Hu-Tang, consisting of Ephedra sinica, Prunus armeniaca, Glycyrrhiza uralensis, Morus alba, Asarum heterotropoide, Zingiber officinale is a famous prescription used as an antitussive and antiasthmatic agent in the traditional Chinese medicines.On the basis of research on the efficacy of Wu-Hu-Tang, we carried out the investigation of its in vivo effective substances, and obtained some interesting results:.1),The 19 in vivo chemical constituents were identified after Wu-Hu-Tang was orally administered to rats, and they were glycyrrhizic acid, glycyrrhetinic acid-3-O-b-glucuronide, glycyrrhetinic acid, oxyresveratrol, mulberroside A, N-methylephedrine, ephedrine, pseudoephedrine, phenol, phenethyl alcohol, 3-methylphenol, α-terpineol, 2,6,6-trimethyl-bicyclo(3.1.1)heptan-2,3-diol, terpinol, borneol, 1,4-benzene diol, 4-methoxymethylphenol, 4 -(4-hydroxyphenyl)- 2-butanone, 4-hydroxy-α-methyl-phenylpropanol, 3,4,5-trimethoxyphenol. .2),The in vivo chemical constituents as targets were screened for antiasthmatic agents, and ephedrine, pseudoephedrine, glycyrrhizic acid, glycyrrhetinic acid, oxyresveratrol, borneol, terpineol, phenethyl alcohol were shown to have inhibitory effects against contractions of Guinea pig tracheal smooth muscles induced by histamine. Combination of ephedrine and glycyrrhetinic acid or ephedrine, glycyrrhetinic acid and borneol exhibited synergistic effect. Further study showed that combination of ephedrine and glycyrrhetinic acid synergistically enhanced the relaxant effect of Guinea pig tracheal smooth muscles or the inhibitory effect against contractions of Guinea pig tracheal smooth muscles induced by mixture of phosphoric acid and histamine, but could not enhance the relaxant effect against contractions of Guinea pig tracheal smooth muscles induced by the mixture. .3)、The in vivo experiment showed ephedrine, glycyrrhetinic acid, borneol and terpineol markedly prolonged latent period of asthmatic attack by histamine in Guinea pigs, suggesting that they have antiasthmatic effects;Combination of ephedrine, glycyrrhetinic acid and borneol showed the synergistic effect against asthmatic attack of Guinea pig induced by histamine..4)、The in vivo chemical substances-terpineol, phenethyl alcohol, benzaldehyde, glycyrrhizic acid, borneol as targets were screened for antitussive agents, and terpineol, glycyrrhizic acid, borneol prolonged the latent period of cough and decreased the cough frequency induced by SO2 in a dose-dependent manner, suggesting that those three constituents possessed antitussive effect. However, combination of ephedrine, glycyrrhetinic acid and borneol showed no synergistic effect in the experiment. .The above studies showed that the in vivo antiasthmatic and antitussive substances consist of kinds of chemical constituents derived from different drug materia, and there have synergistic effects among some constituents, which partially demonstrated why the prescription combination of Wu-Hu-Tang is reasonable.
通过对大鼠灌胃五虎汤后血浆、尿、胆汁中化学成分的分析、分离和鉴定,确定进入体内的五虎汤来源成分,并以它们为靶向成分进行镇咳平喘相关药效学研究筛选出有效成分,进而在此基础上通过有效成分组合的药效学实验探讨组方的合理性或简方的可能性。此方法不仅为中药复方体内作用物质基础的研究提供新的思路,而且为临床用药提供直接的理论依据。.
