Glycosylphosphatidylinositol specific phospholipase D (GPI-PLD) cDNA with full length 2.6 Kb (GenBank Accession AY007546) and biological functions has been first cloned from the stromal cells of human bone marrow, its genomic gene structure has been confirmed, these data will be very usefulness for further studying GPI-PLD gene expression and its action in hematopoiesis of human bone marrow. The polymorphism of GPI-PLD gene was identified, the method of quantificationally competitive RT-PCR for GPI-PLD mRNA was build and the changes of GPI-PLD gene expression in liver diseases and leukemia patients were found, our these results showed that GPI-PLD gene are very usefulness for clinic,too. 4 papers have been published in the top scientific journals and 3 graduate students with Master Degree have culured after this grant funds.
用RT-PCR技术从人骨髓基质细胞获得糖基化磷脂酰肌醇特异性磷脂酶D的cDNA,构建成穿梭质粒后感染大肠杆菌,筛选出阳性克隆并测出该克隆基因的全序列,为进一步研究该酶在骨髓造血活动中所起作用打下基础。从克隆的基因制备出探针,分析白血病患者该酶的基因结构,为该病研究开辟新的领域。该基因在真核细胞表达的产物经纯化后可带来经济效益。
