Many molecular events occurred during the processing of colorectal cancer. The emphasis in treating cancer was developing molecular targeted medicine based on the study of cancer gene and restricting cancer gene. The most important thing was to find the key molecular during the colorectal cancer development and evaluate its effects. It was proved that estrogen correlated with colorectal cancer significantly. The C-Abl gene played an important role in the development of cancer by human genome gene sequencing technique based on previous research. Additional research proved the mutation of C-Abl gene may take place by the induction of the estrogen. However, how did the estrogen regulate the expression of C-Abl gene, what role did the C-Abl gene play in the colorectal caner cell inducted by the estrogen? The mechanism was still unknown. This objective of the study was to evaluate the mechanism of C-Abl gene expression inducted by estrogen and the effects of C-Abl gene on the invasion and metastasis of colorectal cancer by siRNA, cDNA, cell invasion,cell migration,cell cycle, apoptosis and in vivo tests, elucidating the mechanism of colorectal cancer development inducted by the estrogen systematically and developing new treatment methods of the cancer.
大肠癌的侵袭、转移贯穿了许多的分子事件的变化,基于对大肠癌癌基因、抑癌基因的认识,开发的分子靶向治疗药物是目前临床治疗大肠癌工作的重点,通过文献回顾证实雌激素与大肠癌的侵袭转移密切相关,前期通过人类全基因组测序技术证实C-Abl的基因突变可能与大肠癌的疾病进展过程中发挥着重要的作用,进一步的研究证实:C-Abl的基因突变可能是通过雌激素的诱导而发挥作用的,但雌激素如何调控C-Abl的表达,C-Abl在雌激素诱导的大肠癌细胞中发挥着怎样的作用及机制尚不清楚。本课题拟采用siRNA、cDNA、染色质免疫共沉淀、细胞侵袭迁移、细胞周期、凋亡以及体内实验等方法,探讨雌激素诱导C-Abl表达的转录调控机制及C-Abl在雌激素的诱导下对大肠癌侵袭转移的影响,为系统阐述雌激素诱导大肠癌侵袭转移的机制提供理论依据
