Pseudomonas aeruginosa is a ubiquitous gram-negative rod responsible for acute and chronic lung infections wih severe antimcribial resistance. Innate immunity, specifically, professional phagocytic cells as a key determinant in the ability of the host to control P. aeruginosa infection. Keratinocyte growth factor-2 ( KGF-2), a potent alveolar epithelial cell mitogen, could promote the proliferation, migration and differentiation of epithelial cells. Our preliminary study found that KGF-2 could enhance the phagocytosis of macrophage cells, and improve the survival of P.aeruginosa infected mice, but the mechanism remains unclear. The project intends to stimulate macrophages that pretreated with KGF-2 or not by pseudomonas aeruginosa, and to study its influences on macrophage phagocytosis and inflammation; explore the mechanism of KGF-2 on the macrophage phagocytosis and inflammation; establish P.aeruginosa lung infection model, assess the impact of KGF-2 on the inflammatory response of P. aeruginosa induced lung infection, as to clarify the role and mechanism of KGF-2 on the regulation of P. aeruginosa induced lung infection. This study will provide theoretical basis for the clinical application of KGF-2.
铜绿假单胞菌是重要的导致急慢性肺部感染的机会致病性革兰氏阴性杆菌,耐药现象严峻,巨噬细胞为代表的固有免疫在控制绿脓感染中发挥了重要的作用。角质细胞生长因子-2(KGF-2)作用于上皮细胞表面的酪氨酸激酶受体FGFR2-IIIb或FGFR1-IIIb,促进上皮细胞的增殖、移行和分化。我们前期研究发现巨噬细胞表达FGFR2-IIIb受体,KGF-2可增强巨噬细胞的吞噬功能,改善铜绿假单胞菌肺部感染小鼠的生存率,但其作用机制尚不清楚。本项目拟通过体外铜绿假单胞菌感染巨噬细胞,研究KGF-2对巨噬细胞吞噬功能和炎症反应的影响,探索KGF-2调控巨噬细胞吞噬功能和炎症反应的作用机制;体内建立铜绿假单胞菌肺部感染模型,KGF-2预处理对铜绿假单胞菌肺部感染中的作用。本研究将为KGF-2应用于铜绿假单胞菌肺部感染提供前期的理论基础。
铜绿假单胞菌是重要的导致急慢性肺部感染的机会致病性革兰氏阴性杆菌,耐药现象严峻 ,巨噬细胞为代表的固有免疫在控制绿脓感染中发挥了重要的作用。角质细胞生长因子-2 (KGF-2)作用于上皮细胞表面的酪氨酸激酶受体FGFR2-IIIb或FGFR1-IIIb,促进上皮细 胞的增殖、移行和分化。我们研究发现KGF-2无论是预防给药还是治疗给药,均能够显著改善铜绿假单胞菌肺部感染小鼠的死亡率,并且降低肺内炎症反应,减少肺水含量,降低肺内炎症因子的释放水平减轻肺损伤,通过增加肺泡内巨噬细胞及中心粒细胞的数量,增强肺组织局部免疫功能,进而增加肺组织局部细菌吞噬,减少细菌入血。另外,我们发现小鼠巨噬细胞表达FGFR2-IIIb受体,KGF-2可增强巨噬细胞的吞噬功能。本研究证实KGF-2预防给药及即时给药均能改善铜绿假单胞菌肺部感染小鼠的生存及肺损伤,可能与KGF-2增强肺内局部吞噬细胞免疫功能,进而减轻炎症反应有关,改善了肺组织修复。并且我们初步发现KGF-2可增强巨噬细胞吞噬功能。本研究为KGF-2 应用于铜绿假单胞菌肺部感染提供前期的理论基础。
