The underlying mechanism of the angiogenesis, tumor invasion and metastasis are important to the clinical therapy of pancreatic cancer. Our previous study had found that Reg3α (regenerating islet-derived protein 3α) was highly expressed in pancreatic cancer tissues, which was related to the expression level of Angiopoientin-1 and angiogenesis in pancreatic cancer. And using of recombinant Reg3α, the proliferation and migration of vessel epithelial cells were improved. Reg3α might induce its receptor EXTL3 translocation into nucleus, thereby increasing the expression of Angiopoietin-1. Moreover, interference of Angiopoietin-1 pathway could reduce the pro-angiogenesis effect of Reg3α. Therefore, Reg3α could strongly associated with the angiogenesis in pancreatic cancer via regulating Angiopoietin-1 signal. This study would further investigate the relationship of Reg3α and angiogenesis in pancreatic cancer according to clinical study, in vitro and in vivo experiments. Further exploration of the biological effect of Reg3α on angiogenesis should provide benefit to the diagnosis and treatment of pancreatic cancer.
胰腺癌新生血管形成及侵袭转移的分子调控机制是目前临床医学研究的热点和难点。本课题前期研究发现Reg3α在胰腺癌临床样本中显著高表达,并与Angiopoietin-1的表达水平、新生血管形成及肿瘤侵袭转移正相关;Reg3α重组蛋白刺激能促进血管内皮细胞增殖、迁移,诱导其受体EXTL3活化入核及Angiopoietin-1表达增加;干扰Angiopoietin-1可有效降低Reg3α促新生血管形成的生物学功能。由此,本课题推测Reg3α可通过促进Angiopoietin-1转录,介导胰腺癌新生血管形成和侵袭转移。本课题拟从临床样本验证、细胞及动物实验多个层面,进一步证实Reg3α与胰腺癌临床特征及预后的相关性,明确其生物学功能及通过与EXTL3结合促进Angiopoietin-1转录的作用,旨在阐明Reg3α在胰腺癌中异常表达的调控机制,为临床诊断、预防及治疗胰腺癌提供新的分子靶标和研究思路。
胰腺癌是一种侵袭性很强、恶性程度极高的消化道实体肿瘤由于早期诊断率低且初期即发生局部淋巴结及远处转移,外科手术后也易发生侵袭转移而造成复发,胰腺癌是预后最差的恶性肿瘤之一,5年生存率仅为1% - 4%,寻找有效抑制胰腺癌侵袭转移的分子靶标一直是当前的研究热点和难点。REG3A(regenerating islet-derived protein 3A)是胰岛再生源蛋白家族成员之一,其在结直肠癌、肝细胞肝癌等多种恶性消化道肿瘤中的表达均异常增高,已被指出可作为促肿瘤增殖因子参与多种恶性肿瘤的发生发展,具有作为胰腺癌临床诊断、治疗分子靶标的潜在可能。但是作为一种广泛调控细胞增殖的再生的蛋白,REG3A在胰腺癌发展过程中的具体调控机制仍未完全清楚。.本研究首先调研了临床胰腺癌患者血清中REG3A表达水平与胰腺癌进展、肿瘤组织REG3A表达水平与肿瘤细胞增殖、迁移的关系,发现REG3A高表达显著影响了胰腺癌进展与细胞生长。为进一步研究REG3A调控胰腺癌细胞生长的具体机制,通过构建表达REG3A的腺病毒载体,在胰腺癌细胞中过表达REG3A;同时利用外源性重组蛋白,在细胞水平研究REG3A对胰腺癌细胞生长的直接刺激作用及具体作用机制。发现REG3A在正常胰腺癌细胞中作用较弱,但在缺血损伤时,REG3A通过EXTL3介导了缺血损伤状态下的胰腺癌细胞保护作用,从而在组织水平中介导了缺血部位的肿瘤细胞生长,促进胰腺癌细胞生长迁移。.本研究首次发现了REG3A在胰腺癌缺血状态下调控肿瘤生长的生物学机制,阐释了REG3A在胰腺癌患者血清中升高的作用机制,为REG3A应用于胰腺癌诊断、治疗提供了潜在的作用靶点。本项目的主要研究结论已撰写论文拟投稿。此外,基于胰腺癌的肿瘤分子诊断,已在项目支持下发表第一作者SCI文章3篇,申请人获得江苏省科教文卫青苗工程资助。
