Phosphatidylinositol-3-OH kinase (PI3K) plays a pivotal role in the proccess of monocyte/macrophage migration during inflammation, however, the activation of PI3K/Akt in turn suppresses inflammation. How macrophages coordinate migration and activation during inflammation, the molecular mechanisms underlying this prossecc have not been characterised. Our results show that knockdown PH domain-containing protein PLEKHQ1 resulted in impaired NF-κB activation and downregulation of pro-inflammatary factors IL-6 and TNFα after LPS stimulation. Whereas, PI3K/Akt signaling was overactivated. Meanwhile, overexpression of PLEKHQ1 inhibited macrophage migration. These data suggest that PLEKHQ1 plays a key role in the regulation of macrophage migration and activation. In this project, we plan to investigate the precise role of PLEKHQ1 in macrophage migration and activation in macrophage cell lines and knock out mice in vivo. We also plan to explore the molecular mechanisam through protomics and biochemical technolgy. This project will reveal the molecular mechanism which balances the migration and activation of macrophage, and provide new insights into the therapy of diseases such as atherosclerosis which are relative to macrophage migration and activation.
单核/巨噬细胞迁移到炎症反应位点的过程中,磷脂酰肌醇3激酶(PI3K)的激活起关键作用,然而PI3K的激活又抑制炎症反应。巨噬细胞如何协调迁移与促炎功能的发挥,其机制还未被揭示。我们的前期结果显示,在巨噬细胞中敲低PH结构域蛋白PLEKHQ1后,LPS激活的NF-κB信号通路减弱,炎症因子IL-6及TNFα的表达下调,但PI3K/Akt通路过度激活。而过表达PLEKHQ1抑制巨噬细胞的迁移。这些结果提示PLEKHQ1是巨噬细胞迁移及激活的关键调控因子。本项目拟采用巨噬细胞系以及PLEKHQ1基因敲除小鼠为研究对象,深入研究PLEKHQ1对巨噬细胞迁移和激活的调控作用。并通过生化和组学方法阐明分子机制。本项目的完成将揭示平衡巨噬细胞迁移与发挥促炎症功能的分子机制,为动脉粥样硬化等与巨噬细胞激活和迁移相关疾病的治疗提供理论线索。
巨噬细胞的迁移和激活是炎症反应过程中两个必要过程。其中磷脂酰激酶PI3K的激活是巨噬细胞迁移所必需的,但PI3K持续的激活又抑制这些细胞的激活,不利于炎症反应的充分发生和病原体的高效清除。巨噬细胞如何协调迁移和激活有序发生,这一过程中的分子机制还不清楚。我们的研究发现,PH结构域蛋白PLEKHQ1(又称PLEKHO2)是协调巨噬细胞迁移和激活的关键分子。PLEKHQ1在巨噬细胞中高表达,并且随着巨噬细胞分化表达上调。PLEKHQ1促进LPS介导的巨噬细胞迁移,并抑制LPS诱导的巨噬细胞激活,进而可以协调巨噬细胞迁移与发挥促炎症反应的平衡。整体水平上,建立了PLEKHQ1基因敲除小鼠,发现该小鼠对LPS诱导的炎性反应更加敏感。分子机制方面,PLEKHQ1通过抑制LPS/TLR4通路下游关键分子TRAF6的激活而抑制LPS诱导的巨噬细胞激活。PLEKHQ1可结合磷脂并促进PI3K/Akt活化促进巨噬细胞迁移。这些结果证明PLEKHQ1是一个协调巨噬细胞迁移与炎症反应平衡的关键分子。
