Diabetic peripheral neuropathy is the one of the main complications of diabetes which affects a lot of patients in the whole world. How to improve the axon regeneration become important. Schwann cells are the principal glia of the peripheral nervous system (PNS). There are two types of Schwann cell, myelinating and nonmyelinating. Myelinating Schwann cells wrap around axons of motor and sensory neurons to form the myelin sheath and made axon regeneration. c-jun/Krox2 regulates the differentiation of Schwann cells. P38 MAPK pathway was considered as a upstream signaling pathway. There is no research on the Schwann cells phenotype change in diabetic Diabetic peripheral neuropathy(DPN). Former research has confirmed Jinmaitong have good therapeutic effect in DPN. Experiments in vitro have proved the Jinmaiton could improve the MCV of DPN rats,suppressed apoptosis of SCs and promote the expression of neurotrophic factors et al. Our group is aimed to research the mechanism of Sc differentation in DPN and the Jinmaitong effects on above pathology.
糖尿病周围神经病变(DPN)是困扰糖尿病患者的主要并发症之一。如何促进DPN神经损伤修复成为近年来研究的热点。雪旺细胞(Sc)是修复神经元损伤的主要细胞。周围神经损伤时,雪旺细胞可分化为髓鞘形成表型,表达大量神经营养因子,从而促进轴突再生用。P38MAPK/c-jun/Krox2通路是调节雪旺细胞分化的主要通路之一,目前对于糖尿病周围神经损伤时雪旺细胞表型的变化以及高糖是否可通过P38MAPK/c-jun/Krox20通路导致其表型的改变尚未见报道。而既往研究证实中药筋脉通治疗DPN临床疗效良好,体外试验证实其能够改善DPN大鼠的神经传导速度,抑制雪旺细胞凋亡、促进神经营养因子表达等作用。本课题拟采用DPN大鼠模型及雪旺细胞、脊髓背根神经节细胞共培养体外模型,利用分子生物学技术研究DPN中雪旺细胞基因表型的表达及中药筋脉通是否可通过抑制p38 MAPK通路、调节雪旺细胞基因表型的表达从而促进糖尿病周围神经病变修复。
