Lipoprotein abnormalities is the most common complication of obesity and type 2 diabetes (T2DM). The typical performance of lipoprotein abnormalities is metabolic abnormalities of lipoprotein complex in composition, structure and function. However, it is unclear the composition, abundance of protein and lipid included in lipoprotein complex in the development of obesity and T2DM at systemic level. Based on a prospective population cohort sample, this study will build the dynamic pattern of proteins and lipids in plasma lipoprotein complex (VLDL-LDL-HDL) by proteomic and lipidomic strategy and comprehensively integrate knowledge of the composition, structure, metabolism, and function of lipoprotein complex for revealing the distinct metabolic pathway of lipoprotein complex involved in the development of obesity and T2DM, demonstrating that the causal relationship between lipoprotein abnormities and obesity and T2DM and providing proteins and providing proteins and lipids group for predicting of obesity and T2DM. This study will not only improve the screening of individuals with high-risk for obesity and T2DM, but will also provide new insights into mechanism investigation and development of population intervention program for this disease.
脂代谢紊乱是肥胖和2型糖尿病(T2DM)最常见的并发症,其典型表现为血浆脂蛋白复合物组成、结构和功能的代谢异常。目前,对于肥胖和T2DM发生发展过程中血浆脂蛋白复合物包含的蛋白质和脂质在系统层面上发生的变化尚不清楚。为此,我们拟在前瞻性人群队列样本基础上,运用蛋白质组和脂质组技术绘制肥胖和T2DM发生发展过程中血浆脂蛋白复合物(VLDL-LDL-HDL)的蛋白质和脂质的动态变化图谱,全面解析血浆脂蛋白复合物的组成、结构、代谢和功能,揭示血浆脂蛋白复合物在肥胖和T2DM发生发展过程中参与的重要代谢途径,阐明血浆脂蛋白复合物异常与肥胖和T2DM发生发展的因果关系,提供在肥胖和T2DM发生发展过程中具有预警作用的蛋白质和脂质组合。本研究的开展将有助于筛选肥胖和T2DM高风险人群,为后续该病发生发展的相关机理研究和人群干预方案的制定提供新的线索和思路。
