通过厚皮甜瓜雄花、两性花同株(aaGG)与雌、雄异花同株(AAGG)基因型间杂交,获得1000株F2的构图群体,采用BSA策略和PCR为基础的标记技术,对控制厚皮甜瓜雌、雄异花同株花性型基因A进行分子标记和精细定位,并构图甜瓜的高饱和连锁图谱。将筛选出的与A基紧密连锁的标记用于回交转育的标记辅助选择,培育"AAGG"基因型自交系,筛选强优势组合。.通过对A基因的精细构图,将为A基因的图位克隆奠定基础;在通过回交培育AAGG型自交系的同时,也就培育了轮回亲本"aaGG"的近等基因系"AAGG",为从表达水平克隆与花性型相关的基因奠定了基础;A基因的克隆,可以从分子水平研究葫芦科雌花和两性花中雄蕊的发生与发育机理。.高饱和连锁图谱将为甜瓜结构基因组学和葫芦科比较基因组学研究提供必备的工具。
