Mandibular and TMJ reconstruction is a main problem in maxillofacial surgery. Tissue engineering provides an alternative method to reconstruct the candylar defect. Allogeneic bone has a similar shape and biological properties. Freeze-dried allogenic bone can be used for scaffolds for bone engineering。This study will use human umbilical cord mesenchymal stem cells (hUCMSCs) as basic cells, firstly induce hUCMSCs to differentiate into osteoclasts, and prepare thermo-sensitive Chitosan gel at the same time, then use Chitosan gel as the support to load directional differentiation cells and culture in vitro, observe the feasibility of Chitosan thermo-sensitive Chitosan gel as an injection type carrier of cells; secondly made certain pore in lyophilized allogeneic condyle, then Inject the chitosan gel with directional differentiation cells to the pore and cultured in vitro, observe characteristics of cell differentiation; Finally Through the vivo experiment to confirm the possibility of implant the gel loaded with differentiated cells to pores in allogeneic bone can construct tissue engineered condyle. The study of whether thermo-sensitive Chitosan gel loaded with differentiated hUCMSCs cells as seed cells and allognic candyle as scaffolds can construct tissue engineered condyle, will provide important experiment basis for improve the transplanting success rate of allogeneic bone, and then provide theoretical support to promote the clinical application.
多年来颞下颌关节重建一直是颌面外科的一大难题。组织工程为解决这一难题提供可能,成为国内外学者研究的热点。同种异体骨与自体骨有相似的生物学特性及外形,是较佳的生物支架材料。本课题拟将人脐带间充质干细胞(hUCMSCs)向成骨、成软骨细胞方向分化后,同时制备壳聚糖温敏凝胶作为细胞载体,将壳聚糖凝胶作为支架加载定向分化细胞体外培养,观察壳聚糖温敏凝胶作为细胞的可注射型载体的可行性;将冻干的同种异体下颌骨髁突制成一定的孔隙,加载有定向分化细胞的壳聚糖凝胶注射于离体同种异体骨孔隙内体外培养,观察异体骨支架、壳聚糖凝胶及诱导分化的hUCMSCs之间相容性;最后通过动物体内实验验证其构建组织工程髁突的可能性。研究同种异体骨为支架、壳聚糖温敏凝胶加载双向诱导分化的hUCMSCs种子细胞能否构建组织工程髁突,将为提高同种异体骨移植髁突重建的成功率提供重要的实验依据
颞下颌关节修复重建时口腔颌面外科临床上一个亟待解决的难题,本项目针对这一临床问题,采用同种异体骨作为支架材料,人脐带间充质干细胞作为种子细胞,以壳聚糖凝胶作为细胞载体,研究发现从人脐带分离获得间充质干细胞,在体外可稳定传代培养,具有多向分化能力,生物学性能稳定。通过诱导,能够向成骨软骨细胞分化。同时发现同种异体骨支架、壳聚糖凝胶及诱导分化的hUCMSCs之间具有良好的生物相容性;并通过动物体内实验验证其构建组织工程髁突的可行性。为提高同种异体骨移植髁突重建的成功率提供重要的实验依据。.本项目研究内容进展顺利,已发表和接受相关SCI论文4篇,中文核心源期刊9篇,培养博士研究生5名,硕士研究生1名。获得陕西省科技进步二等奖1项。
