Phenolic compounds can interact with other nutrient components and affect their bioavailability, which lead to great difference in vivo and in vitro. Based on composition analysis of naked oats phenolic compounds and regulation of the intestinal micro-ecology, the metabolic transformation of oats active ingredients, changes of gut microbiota and evaluation of activity will be focused on to explain the interaction of phenolic compounds with beta glucan and gut microbiota, which will provide foundation for their physiological function..The forms of main phenolic compounds and beta glucan from naked oats will be studied by in vitro simulated gastrointestinal digestion and colonic fermentation. The digestion of biological availability and absorption mechanism of naked oat wheat phenolic compounds will be evaluated by Caco-2 cell monolayer model. The interaction mechanism of active components (i.e., naked oats phenolic compounds and beta- glucan) with gut microbiota will be explored. The antioxidant activity and oxidative stress regulation mechanism in endothelial cells of different forms of naked oat free phenolic compounds and phenolic beta-glucan polymers will be compared. This digestion, absorption and metabolism mechanism of nutrition components in naked oats will be investigated, which will be helpful to evaluate their bioavailability and can provide solution for the processing method.
酚类化合物可与其他营养组分互作影响其生物利用度,导致其体内外活性差异较大。基于裸燕麦酚类化合物组成分析和调节肠道微生态等研究,本课题将从裸燕麦主要活性成分代谢转化、肠道菌群变化以及活性组分评价三方面探讨酚类化合物和β-葡聚糖等活性组分与肠道微生物互作关系,为其生理功效阐明提供依据。.拟以裸燕麦酚类化合物和β-葡聚糖为对象,采用体外模拟胃肠消化和结肠微生物发酵探讨裸燕麦特征性酚类化合物在不同消化阶段的存在形式;以Caco-2单层细胞模型评价为指针,探讨裸燕麦酚类化合物在各消化阶段的生物利用度和吸收机制;探究以裸燕麦酚类化合物与β-葡聚糖等活性组分与肠道微生物互作效应;比较裸燕麦游离态酚类化合物、酚类-β-葡聚糖聚合物等不同存在形式的酚类化合物的抗氧化活性及内皮细胞氧化应激调控机制。本项目有助于深入理解裸燕麦营养组分的消化、吸收、代谢机制,合理评价其生物利用度,为改进裸燕麦加工方式提供依据。
