3个人胃癌差异表达基因全长cDNA克隆及染色体定位研究

Abstract: Gene gcys-20, a differentical display gene of gastric carcinoma, is localized at from 29-3 to 29-8 on chromosome 14. Gcys-1 is localized at 3q42 on chromosome, and its access number in GeneBank is AF071052. Gcys-18 is a homoeotic gene of gene erk, and its access number in GeneBank is AF071056. Its chromosome location is just as same as gene erk's. We testified that the mammary cancer antigen correlative gene brcaa1 was higher expressed in gastric carcinoma and it was lower expressed in normal gastric mucosa. Gene brcaa1 is localized at 1q42.1-43 on chromosome and its access number in GeneBank is AF208045. Our research is a foundation of clarifying the function of these genes in occurring and developing of gastric carcinoma.

应用改进的差异展示技术从胃癌7901与胃粘膜GES－1细胞株中获取了154个差异表达基因片危擞迷咏患际醵?8 例胃癌及100例胃癌前变粘膜检测，筛选出3个特异性好在胃癌及癌前变粘膜中组合检出率92.3%的新基因片段，拟筛选胃癌cDNA片段，设计引物，运用快速扩增cDNA末端技术分别获取5"端与3"端cDNA序列，以全长克隆作探针进行染色体定位。
