Utilizing the techniques of DDRT-PCR and suppress subtract hybridziation,a number of differentially expressed genes were isolated from the lung cancer cells induced by radon or αparticle exposure .The mRNA expression changes were confirmed in some of these lung cancer-related genes by Northern (or reverse) blot hybridization. The 1.3kb full-length cDNA of a unknown-function gene LCA295 were cloned by RACE technique. This transcript is composed of 3 exons and 2 introns. Its encoding produt is a 75AA of small molecule protein. This protein is highly conserved from plant to human.Our experiment indicated that the cellular localization of LCA295 protein was gradually condensed into nuclei and a "central line" was formed in late M phase.In this project, a novel DNA-PKcs binding protein, cyclin T2 was identified.Cyclin T2 can binding with RNA polymerase, involves in the regulation of some genes expression.This result suggests that the changed expression of DNA-PKcs could relate to the cellular transformation through the action of gene expression regulation.Another isolated gene was confirmed to be the gene encoding human Bcl-2 modifying factor (BMF).
居室内氡暴露所致肺癌已构成一个重要的公众健康问题,对其危险度评价具有意义。本研究拟采用已在云锡矿工肺癌研究中所得差异基因片段,验证其是否在受高氡浓度暴露的窑洞内长期生活的居民肺癌标本中表达差异,获得相关基因全长序列,通过结构与功能研究了解其生物学意义,初步阐明此基因在该类肺癌中的作用机制,寻找用于诊断或预后特异性标记物
