Elite parents have outstanding contributions to crop breeding programs, therefore, it can provide scientific basis on molecular design breeding by realizing evolusion trend and effects of important chromosome zones in elite parents. In previous study, key chromosome zones of Guichao 2, a rice elite parent, were identified by genome-wide screening based on SSR. However, further research into transfer patterns and effects of these chromosome zones is needed. SNP genotyping of key chromosome zones will be performed by allele-specific PCR (AS-PCR) to find out allele distribution; agronomic characters investigation and pedigree analysis will be carried out to identify transfer patterns and effects of key chromosome. Furthermore, the correlations between SNP loci and phenotypic variation will be analyzed by using association analysis.These research will be bound to benefit rice molecular assisted selection, acquisition of markers close linked with phenotypes, gene clone and functional analyzing, and so on.
骨干亲本在育种实践中有突出贡献,认识骨干亲本关键基因组区段的演变趋势及效应可为基于全基因组的分子设计育种提供科学依据。在前期研究中,申请人利用SSR全基因组扫描确定了水稻骨干亲本桂朝2号的关键基因组区段,这些区段在衍生品种中的传递和效应还需进一步探讨。本项目拟采用等位基因特异PCR法对关键基因组区段的候选基因进行SNP分型,了解有利等位基因的分布,结合表型鉴定与系谱分析,阐明关键基因组区段的传递规律及效应,为骨干亲本和衍生品种的利用提供科学依据。根据SNP分型结果及表型数据,确定与表型变异相关性较高的SNP位点,构建群体对这些位点进行关联分析,明确SNP对表型变异的贡献,从而开发与性状紧密连锁的分子标记,以期为水稻分子设计育种、候选基因的克隆及功能分析等工作奠定基础。
