Liver fibrosis is a high incidence of the disease,But there is no ideal way to reverse it . So it is an urgently need to find new treatments . The bone marrow mesenchymal stem cells (BMSCs) is a kind of differentiation of pluripotent cells, under certain conditions, can differentiate into hepatocyte-like cells, thereby reversing liver fibrosis . However, its clinical efficacy depend on BMSCs homing and differentiation , SDF-1/CXCR4 axis is a key signaling pathways on BMSCs homing.Theoretically , on one hand Biejiajianwan made the " blooding, Blooding Circulation, stasising" ensure BMSCs the smooth running in the blood , help BMSCs homing , on the other hand depending the " nourishing yin qi and nutring " on improving liver microenvironment protect BMSCs differentiation , regeneration nutrition. Can use Biejiajianwan strengths and multi-target anti- hepatic fibrosis characteristic of stem cells liver fibrosis? Nobody has studied. The project as a starting point, through Biejiajianwan intervention BMSCs induced differentiation experiments and experimental liver fibrosis , liver -specific cell lines (CK-18, ALB, AFP) was observed after BMSCs transplantation and SDF- 1/CXCR4 and other changes in terms of homing and differentiation of BMSCs investigate the effect of liver fibrosis Biejiajianwan treatment of stem cells. in a word ,it sprovide the basis for tem cells combined traditional Chinese medicine .
肝纤维化发病率高,目前无逆转肝纤维化的理想方法。骨髓间充质干细胞(BMSCs)是一种具有多潜能分化的细胞,在一定条件下可分化为肝样细胞,从而逆转和延缓肝纤维化,但BMSCs的临床疗效决定其归巢与分化,而SDF-1/CXCR4轴是影响归巢的关键信号通路。理论上,鳖甲煎丸一方面通过“活血、行血、祛瘀”保证BMSCs在血液中运行通畅,有利于BMSCs归巢,另一方面通过“养肝阴、补气血”改善肝脏微环境,保障BMSCs分化、再生营养。能否利用鳖甲煎丸抗肝纤维化的优势及多靶点作用的特性促进BMSCs抗肝纤维化,目前尚无研究。本项目以此为切入点,通过鳖甲煎丸诱导分化实验及干预BMSCs抗肝纤维化的实验,观察BMSCs移植后肝系细胞特异标志物(CK-18、ALB、AFP)及信号通路SDF-1/CXCR4等的变化,从BMSCs归巢及分化方面探讨鳖甲煎丸对干细胞治疗肝纤维化的影响,为中药联合干细胞治疗提供依据
肝纤维化发病率高,目前无逆转肝纤维化的理想方法。骨髓间充质干细胞(BMSCs)是一种具有多潜能分化的细胞,在一定条件下可分化为肝样细胞,从而逆转和延缓肝纤维化,但BMSCs的临床疗效决定其归巢与分化,而SDF-1/CXCR4轴是影响归巢的关键信号通路。理论上,鳖甲煎丸一方面通过“活血、行血、祛瘀”保证BMSCs在血液中运行通畅,有利于BMSCs归巢,另一方面通过“养肝阴、补血气”改善肝脏微环境,保障BMSCs分化、再生营养。能否利用鳖甲煎丸抗肝纤维化的优势及多靶点作用的特性促进BMSCs抗肝纤维化,目前尚无研究。本项目以此为切入点,通过鳖甲煎丸含药血清作用大鼠骨髓间充质干细胞的体外的诱导实验,与空白对照组及肝细胞生长因子联合成纤维生长因子诱导组进行对照,观察了细胞表达CK-18、AFP、ALB的情况,从而验证鳖甲煎丸含药血清对干细胞向肝细胞转化有促进作用。另一方面,通过制作实验大鼠肝纤维化模型,采用AMD3100阻断SDF/CXCR4信号通路,使用鳖甲煎丸联合骨髓间充质干细胞等进行干预,同时设置空白组、单纯干细胞组及单纯鳖甲煎丸组、模拟组进行对照,观察鳖甲煎丸体外的干预效果及阻断SDF/CXDR4信号轴的后鳖甲煎丸及骨髓干细胞抗纤维化的效果。通过以上两部分实验发现:1.鳖甲煎丸联合干细胞治疗肝纤维化的能明显改善肝纤维大鼠的肝功能、肝脏病理组织,说明中药能作用于干细胞,可用于治疗肝硬化。2.鳖甲煎丸含药血清有促进骨髓间充质干细胞向肝细胞分化的作用,RT-qPCR实验结果可见第28天D组(鳖甲煎丸+ HGF+ FGF-4诱导组)的CK-18表达较其他各组和各时间点明显增高,提示骨髓间充质干细胞在鳖甲煎丸含药血清、肝细胞生长因子和成纤维生长因子-4的共同作用下,表达了CK-18,免疫组化学检测到诱导后的细胞有CK-18、AFP、ALB表达,充分说明鳖甲煎丸含药血清的诱导促使BMSC向肝细胞分化。3.在使用CXCR4的特异拮抗剂后,模拟组与正常组疗效相当,联合组与单纯鳖甲煎丸组的疗效相当,但未加入CXCR4的特异拮抗剂的组别中,各组均有差异,从而推断鳖甲煎丸联合干细胞治疗肝纤维的机制可能通过SDF-1/CRCX4轴起作用。本课题利用SDF-1/CRCX4轴,从BMSCs归巢及分化方面探讨了鳖甲煎丸对干细胞治疗肝纤维化的影响,为中药联合干细胞治疗提供依据。
