Alfalfa (Medicago sativa L.) is a major forage legume and is essential for grasslands and animal husbandry development due to its high quality and vegetative productivity. However, the alfalfa forage with high protein content and low lignin content is highly demanded in our country. During evolution and breeding process, alfalfa stem structure is developed into two different types, solid stem and hollow stem, which directly affects forage quality. But the mechanisms resulting this difference are poorly studied in anatomy, developmental physiology, and metabolic regulation. To elucidate the physiological and molecular basis of the different stem structure formation, we proposed to 1) examine the anatomic structure characters using optical microscope and electron microscope; 2) examine the lignin content and composition; 3) evaluate the forage quality; 4) examine the metabolic regulation and pathway in alfalfa with solid and hollow stem. The knowledge developed in this study will decipher the molecular mechanism for alfalfa stem structure formation and development and may provide new strategies for improving alfalfa forage quality of different stem structures.
随着养殖业的发展需求,对于高消化率、低纤维、低木质素的优质苜蓿品种的需求十分迫切。苜蓿茎特性直接影响苜蓿栽培利用及饲用品质。在生产及研究中发现,苜蓿茎有空心和实心之分,但对不同类型茎解剖结构差异、发育生理及代谢调控过程尚不明晰。本研究拟以实心茎和空心茎苜蓿品种为材料,利用显微镜观察、电镜扫描和代谢组分析等技术,探究苜蓿实心和空心茎生长发育过程中的形态解剖结构动态变化、木质素分布及组成及饲用品质差异,探究苜蓿空心和实心茎生长发育代谢调控途径,为苜蓿茎生长发育分子机制研究奠定基础,为不同茎秆类型苜蓿栽培管理、加工利用及品质改良育种提供新思路和理论依据。
