Jinhuang Powder is a representative formula for Yang Syndrome ulcer of surgery of traditional Chinese Medicine treatment.The clinical efficacy is evaluated.The compatibility embodies the "Treatment of superficial syndrome need hot" theory. Now, the mechanism research on compatibility of Jinhuang Powder was not reported. Because of Yang syndrome was caused by Staphylococcus aureus, which have red, swelling, heat and pain syndrome. This research takes this as the breakthrough point, it study the perspective of molecular mechanism of inflammation of Jinhuang Powder and its compatibility.As the whole, Yang Syndrome Animal Model is the object of study,to use Jinhuang Powder to verify the theory of "Treatment of superficial syndrome need hot ".In the research, real-time fluorescence quantitative (PCR) technology, histopathological observation methods were used. Then, interleukin 1,6,8, TNF- α, IFN content、 expression homogenate TLR2, TLR4, IFN and MYD88 gene in animal models of skin tissue and I κB content were observed,which were to explore the different effect of different compatibility of Jinhuang Powder in warm drugs with other drugs on local inflammatory skin’s microenvironment and TLR2/MyD88/NFκ B channel.This research can guide the clinical medicine medication from the deep research on “Treatment of superficial syndrome need hot " method better and it described the anti-inflammatory mechanism of Jinhuang Powder, that can lay foundations for its further development.
金黄散是中医外科外治阳证疮疡的代表方,临床疗效确切,其配伍体现了"发表不远热"之意,尚未见报道金黄散寒热配伍的机制研究。由于"红、肿、热、痛"的阳症疮疡大都是由金黄色葡萄球菌等引起的急性炎症,本研究以此为切入点,从炎症角度深入研究金黄散寒热配伍的分子作用机制,从整体出发,以阳证疮疡动物模型为研究载体,以金黄散为"发表不远热"法外治阳证疮疡的代表,采用实时荧光定量PCR技术、病理组织形态学观察等研究手段,观察金黄散中温药与其他药物的不同配伍外敷阳证疮疡模型动物后对局部炎症皮肤微环境中白介素1,6,8、TNF-α、IFN及模型动物皮肤组织匀浆TLR2、TLR4、MYD88基因表达及IκB含量变化,探讨金黄散寒热配伍对局部炎症皮肤微环境及TLR2/MyD88/NFκB通路的不同影响,揭示"发表不远热"法的科学内涵从而更好地指导临床用药,并阐释金黄散抗炎作用机制,为其开发利用奠定基础。
