Iridoviruses are large icosahedral cytoplasmic DNA viruses that have been implicated in more than 100 fish species infections and currently threaten the aquaculture industry, causing great economic losses in China. Recently, many host-derived proteins (HDP) on the virus particles were characterized and played important roles in virus infection. However, the roles of host-derived protein on the iridovirus (tiger frog virus) particles in virus infection remain largely uncharacterized. In the present study, a host-derived protein (caveolin-1) on the iridovirus (tiger frog virus, TFV) particles and its roles in virus invasion, assembly and budding are investigated. The host-derived proteins on the TFV viron are identified using the two-dimensional electrophoresis. In order to characterize the function of HDP caveolin-1, the caveolin-1 phosphorylated form and location on the viron, the interaction of caveolin-1 with virus structure protein, and the roles of HDP caveolin-1 in the virus infection and viral immune evasion are detected. We hope our work may help further the understanding of the iridovirus infection and immune evasion.
虹彩病毒是我国水产养殖鱼类重要的病毒性病原之一,每年对水产养殖业都造成了巨大的经济损失。因此,在我国开展虹彩病毒相关研究尤其具有重大的现实意义。近年来,研究发现RNA病毒粒子中携带有大量源于宿主的蛋白(HDP),并且这些蛋白能协助病毒的感染。本课题拟开展虹彩病毒粒子中HDP在病毒感染过程中的作用研究。以虹彩病毒TFV为研究对象,鉴定病毒粒子中携带源于宿主蛋白,以HDP caveolin-1为切入点,阐明虹彩病毒粒子携带HDP caveolin-1蛋白的功能形式和定位、虹彩病毒结构蛋白与HDP caveolin-1蛋白的互作机制、HDP caveolin-1蛋白在病毒感染和调控病毒免疫逃避过程中的作用等三方面研究工作,揭示出虹彩病毒TFV粒子中HDP caveolin-1蛋白的功能,为虹彩病毒其他HDP的功能研究提供借鉴,更为阐明病毒致病机理和免疫逃逸机制提供分子基础。
