: Human leukemia is a common hematopoietic malignant tumor, and its etiology and mechanism remain unknown so far.Our previous studies demonstrated that human leukemic cells contain novel retrovirus. In this study, we successfully isolated and cloned the full-length genome of a novel human retrovirus from primary leukemic cells from leukemia patients using PCR, RT-PCR, RACE, DNA molecular cloning, bioinformatics, transfection assay and other methods. Sequence analysis showed that: the proviral DNA genome consists of 8824 bp, and its genomic structure is5`-LTR-UTR-GAG-POL-ENV-3`LTR; the viral RNA genome is 8400 bp in size, and its genomic structure is 5`-RU5-UTR-GAG-POL-ENV-U3R-POLYA-3`. Alignment analysis and transfection results suggest that this novel retrovirus may belong to type C oncogenic retrovirus. Isolation and identification of the novel retrovirus genome might be of great importance to elucidate the etiology and mechanism of human leukemia.
