Stalk rot, one of the most important and common disease of corn, is becoming increasingly severe recent years, and has stong negative impacts on corn production. The resource of antigen and disease resistant germplasm in maize is limited, thus detecting new resistance genes and creating new disease-resistant germplasm are urgently required. On the basis of massive previous works, in the present study we are going to analyse the functional traits of ZmNBS55Q gene, which is a candidate gene that enhance resistance of corn to stalk rot. ZmNBS55Q gene will be transformed to the inbred lines Chang7-2 (highly susceptible to stalk rot) and Qi319 (resistance to stalk rot), and the transgenic strains with gene over-expression and inferrence expression are expected to be obtained and verified by molecular means. We will subsequently assess the abilitiy of these transgenic strains, verify the function of ZmNBS55Q on disease resistance and clarify the possible mechanism, and screen the new transgenic corn germplasm with resistance to stalk rot. So far, little information is available about the functional analysis and application of ZmNBS55Q both here and abroad. Therefore, this project is significant in both theory and application in detecting new corn stalk rot resistance genes, clarifing the possible mechanisim of corn stalk rot resistance, and cultivating new germplasm resource of disease-resistant genes of maize.
玉米茎腐病是近年来日趋严重的一种重要玉米病害,严重影响玉米生产,但抗源和抗病种质资源贫乏,迫切需要发掘新的抗性基因和创建新的抗病种质。本研究在前期大量研究工作的基础上对获得的玉米抗禾生腐霉菌茎腐病候选基因ZmNBS55Q进行功能分析,拟通过ZmNBS55Q基因对高感茎腐病骨干自交系昌7-2和高抗自交系齐319遗传转化分别获得过表达和干涉表达转基因株并进行分子验证,分析转基因株对茎腐病抗性变化,验证候选基因ZmNBS56Q的抗病功能,初步分析ZmNBS55Q基因抗病的作用机制,筛选出抗茎腐病转基因玉米新种质。该基因的功能分析和应用研究国内外未见报道。因此,该项目研究对发掘新的茎腐病抗性基因,揭示ZmNBS55Q基因抗茎腐病机理,创建抗病新种质新材料,培育抗茎腐病玉米新品种具有重要的理论意义和应用价值。
