Jellyfish (Rhopilema esculentum) as a representative species of Cnidaria, Scyphozoa, has become an important sea catching and cultural animal because of abundant nourishment and medicinal properties. During the breeding process in our previous research, it has been observed that jellyfish has significant difference in body color, including yellow, milky, blue and mulberry. It is common that color differences exists in marine animals, and color diversity has been proved to be significant interrelated with other traits, but the mechanism of color formation remains to be verified..This study was designed for color diversity of jellyfish, utilizing high through-put sequencing technology to conduct comparative transcriptomic analysis in order to obtain differential gene and polymorphic molecular markers underlying color trait of jellyfish. Jellyfish full-sib families were designed to be built aiming at color divergence, taking advances of polymorphic markers obtained from comparative transcriptome analysis, genetic linkage map would be constructed of jellyfish color trait. Linkage analysis and association analysis would then be carried out on the basis of genetic map to acquire candidate QTL or genes significant linkage with color trait. Then candidate genes and differential genes would be cloned and analyzed for structure and molecular function. Ultimately, gene expression of targeted genes in different colored jellyfish would be examined to confirm the reliability of the result for body color gene detection, qualified genes would be regarded to be responsible for color diversity formation in jellyfish.
海蜇(Rhopilema esculentum)是刺细胞动物门钵水母纲的代表物种,因丰富的营养和药用价值成为渔业中重要的海捕和海水增养殖物种。在苗种繁育过程中,观察到食用海蜇表现出黄色、乳白色、蓝色和紫红色四种具有显著差异的表型特征。颜色差异在海洋动物中普遍存在,已有研究也证明个体颜色与一些生长性状具有显著关联,但颜色形成的生物学机制尚未得到证实。.本项目以不同颜色的海蜇为实验材料,基于高通量测序技术对海蜇进行比较转录组学分析,对不同颜色海蜇的转录组进行差异基因筛查并获得大量分子标记。基于颜色差异构建多个海蜇全同胞家系,以转录组分子标记为基础,构建基于海蜇颜色性状的连锁图谱,对调控海蜇颜色形成的基因进行连锁和关联分析,获得与海蜇颜色性状紧密连锁的候选QTL位点或基因。对候选和差异基因进行克隆,并进行结构和功能分析。检测不同颜色个体中目的基因表达差异,最终获得海蜇颜色性状相关基因。
