Gynura bicolor D.C, a special chinese vegetable named "Hong-Jun", is easy to be deteriorated on its quality, and there is almost no postharvest technique and its mechanism research on the vegetable. The applicant's previous research showed that Gynura bicolor D.C treated with 0.50ppm 1-MCP for 12 h at room temperature could effectively improve its defense capabilities by improving the activity of antioxidant enzymes and significantly keep Gynura bicolor D.C quality through delaying senescence. However, its in-depth regulation mechanism is unknown. This project investigates the regulatory mechanism of 1-MCP on antioxidant enzymes such as SOD of Gynura bicolor D.C based on the proteomics technology and combined with the determination of quality, physiological and biochemical changes and adopting Real-time RT-PCR technology. The gene and protein expression modes of antioxidant enzyme- - SOD in Gynura bicolor D.C are expected to obtain, and further regulatory mechanism of 1-MCP on Gynura bicolor D.C postharvest antioxidant enzymes will be clarified through correlation analyses among gene expression, protein expression, physiological and quality changes. 3-4 research articles (including SCI 2) will be published, and graduates students trained 1-2 during research period.
紫背天葵这一中国特色的"红军菜"其采后保鲜技术和相关机理研究鲜见。前期研究表明,室温条件下采后立即施用0.50ppm 1-MCP熏蒸可有效提高其抗氧化酶活性以提高自身防御能力,显著抑制紫背天葵采后品质劣变,延缓衰老,但其深入的调控机理不明。 本项目应用蛋白质组学技术并结合品质、生理生化和实时荧光定量PCR分析,研究1-MCP对紫背天葵采后抗氧化酶的调控作用,以期获得1-MCP处理后紫背天葵抗氧化酶基因表达和蛋白表达情况,通过与生理生化和品质变化相结合的相关性分析,深入阐明1-MCP对紫背天葵采后以SOD为代表抗氧化酶的调控机理。 拟发表相关研究学术论文3-4篇(其中SCI文章2篇),培养研究生1-2名。
1-MCP对果蔬采后的保鲜机制研究开展较多,但大都局限在单一层面。前期研究表明,室温条件下采后立即施用0.50ppm 1-MCP熏蒸12h可有效提高其抗氧化酶活性以提高自身防御能力,显著抑制紫背天葵采后品质劣变,延缓衰老,但其深入的调控机理不明。本项目基于1-MCP处理对紫背天葵生理生化、抗氧化酶蛋白含量变化及其基因表达变化层面开展研究,从不同层面以及各层面间相关性分析,以期获得更全面的1-MCP调控机制。. 本项目的主要成果包括:1-MCP可显著降低呼吸强度,提高SOD等抗氧化酶的活性,并有效保持叶绿素、花青素含量,显著抑制品质劣变;整个贮藏期间,1-MCP显著提高了SOD酶蛋白的含量,且1-MCP对调控紫背天葵蛋白质水平的调节非常复杂,涉及到调控因子蛋白、碳代谢相关蛋白、抗氧化酶蛋白等;基于蛋白质组学的研究结果建立了1-MCP延缓紫背天葵衰老的调节机制图;克隆了紫背天葵SOD基因,对SOD蛋白进行结构预测,SOD荧光表达结果提示1-MCP的上调作用主要为贮藏前中期。根据以上结果明确,1-MCP可通过提高SOD基因的表达,增加SOD蛋白含量和提高SOD酶活性,达到增强紫背天葵抗氧化能力,延缓衰老的作用。. 本项目的研究结果为1-MCP对紫背天葵调控机制的深入研究和控制提供了基础,同时为其他园艺产品采后施用1-MCP保鲜研究提供理论和技术支持。
