The excessive fat deposition caused obesity in body. For human and mammals, the obesity had important role in the development of chronic diseases, such as high blood pressure, cardiovascular disease, cancer, diabetes, etc. Lipogenesis is a complicated process,the profound research on the regulation mechanism of the lipogenesis and obesity would provided the important theoretical basis for the prevention and treatment of those diseases. FSH levels had obviously changed in mammals with ovarian failure and perimenopausal women, and it was reported that fat deposition was regulated by FSH in mouse, but the molecular mechanism have not be known clearly. In this project, the ovariectomy mice were selected. From the molecular,cellular tissue and body levels, the research were comprehensive and in-depth performed to clarify the molecular mechanism of FSH regulating the lipogenesis and obesity, and reveal the relationship between FSH and the obesity of mammals with ovarian failure. The results would provide the theoretical foundation for the obesity and new ideas for the prevention and treatment of obesity in perimenopausal women.
机体内脂肪过度沉积造成了肥胖,哺乳动物和人类高血压、高血脂、心血管疾病、某些癌症和糖尿病的发生都与肥胖相关。脂肪沉积是一个复杂的过程,深入了解脂肪沉积和肥胖发生的分子调控机制将为上述疾病的预防和治疗提供重要的理论依据。 卵巢功能衰退的雌性哺乳动物和围绝经期肥胖妇女体内的FSH水平发生了明显的变化,研究表明FSH能够促进小鼠机体脂肪的沉积,但其分子机理目前还不甚明了。因此,本项目以去卵巢小鼠为实验对象,从分子、细胞、组织和机体水平展开深入系统的研究,以期阐明FSH在哺乳动物卵巢功能衰退后脂肪沉积和肥胖发生中的作用和分子机理,揭示FSH与哺乳动物卵巢功能衰退后肥胖发生之间的关系,为围绝经期妇女的肥胖的研究提供理论基础,并为预防和临床治疗提供新的思路。
