Cervical cancer is one of the mostly common gynecologic malignant tumors. It is very necessary to seek ultrasonic non-invasive treatment for cervical cancer. The non-lethal dose of focused ultrasound is one of the important biological basis for tumor treatment. Our research showed that related proteins of endoplasmic reticulum play key roles in the occurrence and development of cervical cancer. Also autophagy related proteins were closely related to the development of tumor..However, the mechanism of endoplasmic reticulum reaction and autophagy reaction in cervical cancer cells has not reported. Based on these theories and preliminary results, it can provide a new theoretical basis for ultrasound treatment in the occurrence and development of cervical cancer cells..Our research will assess the effect of focused ultrasound on cervical cancer cell line. The effects of focused ultrasound on cervical cancer cells were detected by different doses of non-lethal focused ultrasound. We will measure individual endoplasmic reticulum reaction and autophagy reaction of cervical cancer cell. Also we plan to assess the regulation effect of calcium concentration and the possible mechanism of GRP78 induced by focused ultrasound. To explore the effect of focused ultrasound on the cell by the endoplasmic reticulum pathway, and autophagy pathway, our project will provide new clues to study the effects and mechanism of focused ultrasound for exploring a novel anti-tumor treatment.
子宫颈癌是常见的妇科恶性肿瘤,寻求肿瘤超声无创治疗是目前全球研究热点。目前认为低强度非致死量超声对肿瘤细胞效应是超声治疗肿瘤的重要生物学基础之一。研究表明,宫颈癌的发生、发展与内质网应激相关蛋白有关。自噬相关蛋白与肿瘤的发生发展密切相关。但不同宫颈癌细胞对聚焦超声介导的内质网、自噬反应及相关机制目前尚未见报道。基于这些理论及初步研究结果,可以为超声治疗在宫颈癌细胞发生和发展提供新的理论依据。.本研究以人宫颈癌细胞为研究对象,采用不同剂量非致死性聚焦超声辐照宫颈癌细胞株,检测不同非致死剂量聚焦超声对宫颈癌细胞的影响,了解宫颈癌细胞个体化内质网及自噬反应。探讨内质网应激在超声诱导宫颈癌细胞自噬反应中的作用及机制,并研究超声对宫颈癌细胞的钙浓度的调控,及GRP78在中可能的机制。研究了解内质网途径、自噬途径在在聚焦超声对细胞效应中的作用机制,为聚焦超声杀伤肿瘤细胞的作用机制研究提供新的线索。
子宫颈癌是常见的妇科恶性肿瘤,寻求肿瘤超声无创治疗是目前全球研究热点。目前认为低强度非致死量超声对肿瘤细胞效应是超声治疗肿瘤的重要生物学基础之一。研究表明,自噬相关蛋白与肿瘤的发生发展密切相关。但宫颈癌细胞对聚焦超声介导自噬反应及相关机制目前尚未见报道。。 本研究以人宫颈癌HeLa细胞为研究对象,采用非致死性聚焦超声辐照细胞株,检测宫颈癌细胞自噬反应。研究结果显示,超声诱导宫颈癌HeLa细胞自噬发生,自噬蛋白表达增加,并与超声辐照后时间相关;超声通过自噬途径达到降低细胞活力,自噬抑制剂3MA能抑制超声的杀伤效应,提示诱导宫颈癌 HeLa 细胞内质网应激发生的最小的超声剂量,可诱导宫颈癌细胞发生自噬反应并达到细胞杀伤效应,而自噬抑制剂可以降低超声所导致的细胞损伤。为聚焦超声杀伤肿瘤细胞的作用机制研究提供新的线索。
