Creatine kinase (CK) is one of the key enzymes in cell energy metabolism. Recent studies indicate that the interactions between CK and other proteins have important physiological functions. In a yeast two-hybrid experiment, we found that CK interacts with CC2D1A. CC2D1A is reported to be an activator of NF-κB pathway, and bioinformatics studies indicated that CK also interacts with NF-κB. Our hypothesis is that CK forms an network with CC2D1A and NF-κB, and CK may play an important role in regulating the functions of CC2D1A and NF-κB in this network. In this project, we plan to study the interactions of CK, CC2D1A and NF-κB, and to explore the functions of CK in this network. Our studies will not only clarify the mechanism of interactions between these proteins, but also for the first time, uncover the roles of CK, a key enzyme in energy metabolism, in an important cell signaling pathway of NF-κB. It will shed more light on the physiological and pathological functions of CK.
