Objectives To studies the biological activity, morphology and structure of cell fusion by dendritic cell and squamous carcinoma of human lungs, in order to provide an evidence of experiments and find a new method to prevent and treat human cancer. Methods Dendritic cells of peripheral blood are separated by immunomagnetic bead method (MACS) and incubated in vitro in order to promote it proliferate and differentiate; Fusion cells (FCs) of human dendritic cell and human lung squamous cancer cell are carry out by PEG method; Using immunohistochemistry, light, electron and atomic force microscopy and flowcytometry to observed dendritic cell, human lung squamous carcinoma and fusion cell. In future to study the effect of fusion cell to human peripheral blood in vitro. Results We can get highly purified CD4+DCs with typical morphological and phenotypic features of DC by MACS; 2. We get sufficient typical DCs from CD14+ monocytes induced by combining GM-CSF with IL-4. Monocytes had changed notablely in cytomorphological, cytochemical immuno-stimulating phenotype and immune functions after differentiation; 3. FCs possess the phenotypes of their parental cells; 4. FCs can proliferate in vitro, but which significantly decreased as compared with parental tumor cells; 5. FCs also has a potentiality of stimulating proliferation of mixed T lymphocytes. Conclusions It is enough for doing cell fusion experiment that the human peripheral blood dendritic cell we get by the MACS.PEG method can be used for cell fusion with human peripheral blood dendritic cell and squamous carcinoma of human lungs. The fusion cell can stimulate the T cell proliferate specially, and have some biologic activity of kill the squamous carcinoma of human lungs.
利用细胞分离、纯化、融合技术、蛋白分析及流式细胞技术等观察人肺癌细胞/树突状细胞之融合细胞,以探讨其结构和功能变化;同时观察所提取的融合细胞蛋白对T细胞增殖的影响,以探讨树突状细胞所特异性呈递的肿瘤抗原,为进一步提取肿瘤抗原,参与T细胞的体外特异性活化,体内输入,参与机体特异性抗肿瘤的临床应用提供实验依据。
