AIDS is a major infectious disease threats to life and health, rose rapidly.popular in China, currently about 100 million infected. Chinese medicine is our.unique drug resources, due to the lack of some effective screening method,.resulting in anti-HIV / AIDS seriously lagging behind in the application and.efficacy of Chinese medicine. This topic is intended to establish the role of HIV.and its coreceptor CXCR4 and CCR5 promoter safe drug screening model; luciferase activity by detecting the reporter gene, to observe the impact of drugs on HIV coreceptor; screened with anti-virus the role of traditional Chinese medicine, healthy volunteers oral medicine, the detection of drugs on human peripheral blood T lymphocytes surface CXCR4 and CCR5 expression; virus experiment in vitro analysis filter out Chinese medicine, whether the HIV virus asion and replication inhibition , to verify the effect of antiviral drugs antiviral ingredients; screened by high-performance liquid chromatography analysis, the establishment of a safe and stable anti-HIV / AIDS drug screening system. The system can be extremely sensitive to reflect the drug three Promoter's role, while a high-throughput, fast advantage to provide innovative ideas and methods for screening anti-HIV / AIDS Chinese medicine. 艾滋病是威胁生命健康的重大传染性疾病,在我国的流行呈快速上升趋势,目前国内感染者约为100万。中药是我国特有的药物资源,由于缺乏有的效筛选方法,致使抗HIV/AIDS中药在应用和疗效上严重落后。本课题拟建立作用于HIV 及其辅助受体CXCR4 和CCR5启动子的安全药物筛选模型; 通过检测报告基因萤光素酶活性,观察药物对HIV 及其辅助受体的影响;筛选出具有抗病毒作用的中药,健康志愿者口服中药,检测药物对人外周血T 淋巴细胞表面CXCR4 和CCR5 的表达的影响;体外病毒实验,分析所筛选出的中药,是否对HIV 病毒的侵入和复制有抑制作用, 验证药物的抗病毒效果;高效液相色谱法分析所筛选出的抗病毒药物成分,建立安全稳定的抗HIV/AIDS 药物筛选系统。该系统能够极其敏感的反映药物对三个Promoter的作用,同时具有高通量、快速的优势,为抗HIV/AIDS中药筛选提供了创新性的思路和方法
艾滋病是威胁生命健康的重大传染性疾病,在我国的流行呈快速上升趋势。目前, 全国报告现存活艾滋病病毒(HIV)感染者/AIDS 病人775652例,报告死亡242867例。中药是我国特有的药物资源,随着中医药的发展,中药广泛的应用于艾滋病的治疗,不但能够提高患者生存质量还延缓了疾病的发展。因此,能够在丰富的中药资源中快速的筛选抗HIV/AIDS中药,将为艾滋病的治疗提供新方法。.本课题建立作用于HIV 及其辅助受体CXCR4 和CCR5启动子的安全药物筛选模型。优化并构建了针对HIV辅助受体 CCR5/CXCR4 Promoter 双萤光素酶报告基因载体构建。建立了安全稳定的体外药物筛选系统,对30种中药进行了初步筛选实验,筛选出能了够降低CCR5/CXCR4 Promoter活性的中药主要有:鱼腥草、穿心莲、夏枯草、青蒿、姜黄、雷公藤。选取25名健康志愿者口服中药,并收集志愿者含药血清,验证实验效果,结果显示:中药含药血清穿心莲,夏枯草,姜黄,能够下调 CXCR4 和 CCR5 Promoter 活性。选用假病毒pNL4-3-R-E-载体进行验证药物对HIV复制的抑制作用。ELISA结果显示,中药血清能够降低P24表达。我们从上述中药中提取有效成分:穿心莲内酯、姜黄素,采用双荧光素酶报告基因技术进行验证,结果显示:穿心莲内酯和姜黄素能够显著降低CCR5启动子的活性,而对CXCR4没有明显的抑制作用。本课题建立安全稳定的抗HIV/AIDS 药物筛选系统。该系统能够极其敏感的反映药物对Promoter 的作用,同时具有高通量、快速的优势,为抗HIV/AIDS中药筛选提供了创新性的思路和方法。
