β淀粉样蛋白(β-amyloid protein,Aβ) 代谢异常是老年性痴呆(Alzheimer’s disease,AD)病理机制的中心环节,减少Aβ在脑内的产生、促进Aβ清除、抑制Aβ聚集以降低其神经毒性是AD治疗的新方向。.中医学理论认为AD的基本病机为肾虚痰阻,地黄饮子是补肾化痰的经典方,古代医案和现代临床报道均显示其对AD治疗有很好疗效。前期大量研究证实,地黄饮子能够通过血脑屏障,直接作用于大脑病变组织,能明显抑制APPmRNA的过度表达,显示了其抑制Aβ沉积的良好前景。.本项目以调控Aβ代谢酶活性为策略,采用国际公认的一种转基因小鼠和两种转基因细胞,应用ELISA、Western Blot等技术方法,从Aβ生成和降解途径系统探讨地黄饮子抑制Aβ沉积的作用靶点。本研究进一步系统客观地评价地黄饮子的作用机制,寻找地黄饮子防治AD的作用靶点,为防治AD的经典方新药开发研究奠定基础。
本研究根据Aβ代谢异常这一病理中心环节,着眼Aβ的代谢过程,从Aβ代谢酶入手,通过实验,以两种转基因细胞为靶点,通过观察地黄饮子对Aβ代谢酶的调节作用,研究地黄饮子对Aβ生成和降解过程的影响。. 结果发现地黄饮子对影响Aβ沉积的代谢酶具有抑制作用,对促进Aβ降解的分泌酶具有促进作用。为进一步深入系统地探讨补肾化痰方药地黄饮子防治AD的疗效及机制提供了重要的理论依据,为治疗AD的经典方新药开发研究奠定基础。有利于理解并充实中医的脑髓学说,揭示AD的中医病因病机,为补肾化痰法治疗神经退行性疾病提供了依据。
